摘  要:目的:探讨不同降解时间的聚异丙基丙烯酰胺水凝胶对梗死后心肌组织胶原代谢的影响。方法:合成两种不同理论降解时间的聚异丙基丙烯酰胺水凝胶(Gel A和Gel B)应用于Wistar大鼠心肌梗死模型,Western blot检测心肌组织转化生长因子(TGF)-β1蛋白表达,RT-PCR检测心肌组织Ⅰ、Ⅲ型胶原mRNA水平,心肌组织切片Masson染色观察胶原沉积及计算胶原容积分数。结果:Gel A或Gel B能明显抑制梗死区心肌组织胶原沉积,减少胶原容积分数;有效抑制梗死后增加的Ⅰ/Ⅲ型胶原mRNA比值;但并不能有效抑制梗死后增加的心肌组织TGF-β1蛋白水平。结论:聚异丙基丙烯酰胺水凝胶调节梗死后心肌组织胶原代谢,但这一作用不是通过TGF-β1胶原代谢调节通路实现的。Objective. To assess the influence of Poly N-isopropylacrylamide (PNIPAAm) Gel on collagen metabolism. Methods: Two types of PNIPAAm Gel with different degradation times (defined as Gel A and Gel B) were synthesized. In vivo hydrogel formation and maintenance were observed and confirmed in KM mouse. Left anterior descend of coroanry artery in rats was ligated to make myocardial infarction models in Wistar rats. Western blot was adopted to assay the amount of transforming growth factor (TGF)-β1 protein and PT-PCR was adopted to test the levels of collagen I and collagen Ⅲ mRNA (normalized to β-actin). Masson staining was used to show the collagen deposition in the post-infarcted myocardium. Results: PNIPAAm Gel can significantly inhibit the deposition of collagen and decrease the collagen volume fraction. Western blot test showed that TGF-β1 expression was increased in infarcted myocardium, and remained unchanging after PNIPAAm Gel treatment. Conclusion: The inhibitory effect of PNIPAAm Gel on collagen metabolism is not realized through the TGF-β1 pathway.
