摘 要:利用SSR(简单重复序列)分子标记技术,选取均匀分布于玉米10条染色组的10个标记对20个玉米品种构建DNA指纹库,并研究最佳DNA提取方法和核心引物。结果表明:玉米籽粒快速碱煮法制备的DNA能够满足大批量鲜食玉米杂交种的一致性检测;等位基因数多、PI(多态性信息含量)和MI(标记索引)高且具双亲互补带型的引物被列为指纹库构建的核心引物。一致性检测结果显示:20个品种中的大部分品种R值达50%以上。By means of simple sequence repeat (SSR)molecular marker technology, 10 markers distributed evenly on 10 chromosomes of maize were selected and the DNA fingerprint database of 20 table corn hybrids was constructed. Also the optimal DNA extraction method and core primers were studied. The results showed that the DNAs gained by alkaline lysis of maize grain at 100 °C could meet the consistency check of large quantities of table corn hybrids. The primers having many alleles, high polymorphic information content(PI)and marker index(MI)and being of parent complementary type were regarded as core primers to construct fingerprint database. The consistency check showed that most of the 20 hybrids' R values were above 50%.
