摘 要:采用高效半制备液相色谱法分离,并以13C谱法(13C-NMR)和1H谱法(1H-NMR)确定结构;采用HPLC梯度洗脱进行色谱分离。对不同产地的10批白背叶根药材指纹图谱的相似度进行比较分析。结果表明,分离得到3'-O-甲基鞣花酸-4-O-α-L吡喃鼠李糖苷,纯度>98.50%(面积归一化法);各批药材共建立了15个共有峰,不同产地的白背叶根药材指纹图谱相似度良好。说明高效半制备液相色谱法制备3'-O-甲基鞣花酸-4-O-α-L吡喃鼠李糖苷的方法简单、高效,指纹图谱分析方法稳定性、精密度、重现性较好,用于白背叶根的质量评价切实可行。Semi-preparative HPLC was used to isolate and purify 3'-O-methyl-ellagic acid-4-O-α-L-rhamnopyranoside from the root of Mallotu.1 ape Ita. Its chemical structure was elucidated by ^13C-NMR and ^1H-NMR; 10 batches of the roots of Mallotus ope Ita samples from different sources were analyzed by HPLC gradient elution method. The results showed that the purity of 3'-O -methyl-ellagic acid -4 -O -α-L-rhamnopyranoside reached 98.50% by using area normalization method, 15 common peaks were selected in chromatograms, all the common peaks were separated effectively, and 10 batches of the roots of Mallotus ape Ita fingerprint had good similarity. The results indicated that semi -preparative HPLC was a simple, quick and efficient method to prepare 3' -O-methyl-ellagic acid-4-O-α-L-rhamnopyranoside. Fingerprint analytical method, being with good precision and repeatability, could be useful in the quality control of the root of Mallotus apelta.
