摘  要:【目的】比较拟南芥光敏色素D启动子、基因及其蛋白在黑暗、红光、远红光及蓝光下表达特性。【方法】通过GUS组织染色,半定量RT-PCR及蛋白Western blot检测启动子、基因及其蛋白的表达。【结果】GUS染色显示:子叶中,PHYD基因启动子活性在红光下高于蓝光,且均高于远红光条件;下胚轴中启动子的活性在蓝光条件下最高,远红光条件有较高丰度表达,红光条件下仅在下胚轴上部检测到活性;在根中,蓝光和远红光下生长的幼苗根的上部可检测到启动子的微弱表达,红光下未见表达。在黑暗中生长的幼苗,光敏色素D基因启动子未发生表达。PHYD基因转录和蛋白表达在光下均高于黑暗条件下,不同光质间的表达差异不显著。【结论】在不同光质中,PHYD基因启动子在不同组织的活性存在显著差异;不同光质之间,PHYD基因转录和蛋白表达差异不显著。[Objective] The project focuses on patterns of expression of the Arabidopsis phytochrome D under dark,red light,far-red light and blue light conditions.[Method] The expressive analysis was performed by GUS staining,semi-quantative RT-PCR and western blot respectively.[Result] The expression of PHYD:GUS in different organs of Arabidopsis was regulated in response to different light qualities.There was almost no expression of PHYD promoter in all organs of dark-grown seedlings.The highest expression in cotyledon was examined when seedlings were irradiated with red light,and then in far-red light,in which PHYD promoter was more active than in blue light; In hypocotyls,the promoter expressed at the highest level in blue light,and then in far-red light whereas red-light-induced expression was found only in the upper portion of hypocotyls; For the activities of PHYD promoter in roots,only very low activities were measured in far-red and blue-light-grown seedlings.The expression of PHYD at mRNA and protein level showed light stable under different light conditions.Moreover,results demonstrated that light quality had no effect on the mRNA level and protein abundance of PHYD,except that the lower expression of PHYD mRNA and protein in dark-grown seedlings.[Conclusion] Under different light qualities,the PHYD promoter exhibited different activities significantly in different organs and the expression of PHYD at mRNA and protein level showed light stable under different light conditions.
