摘  要:目的探讨膜联蛋白Ⅱ微卫星的不稳定对前列腺癌早期诊断的作用。方法使用荧光复合扩增的方法对16例前列腺癌组织、16例良性前列腺增生组织及16例正常人血样中的膜联蛋白Ⅱ的三个基因座进行扩增,检测PCR产物,分析其差异与疾病间的关系。结果膜联蛋白Ⅱ的D15S1185,D15S1292两个正常基因座在前列腺癌组织、良性前列腺增生组织以及血液样本中扩增大小无差别;在AN2-di-repeat-NH的二核苷酸重复序列微卫星基因座区域表现差异,以扩增大小134 bp为分界线,前列腺癌组织与正常血液样本存在显著性差异(χ2=9.087,df=1,P=0.003),前列腺癌组织与良性前列腺增生组织之间显著性不明显(χ2=1.997,df=1,P=0.158),良性前列腺增生组织与正常血液样本之间有显著性差异趋势(χ2=3.405,df=1,P=0.065)。结论膜联蛋白Ⅱ二核苷酸微卫星基因座AN2-di-repeat-NH的多态性有可能成为早期前列腺癌的预警指标。Objective To study the effect of annexin Ⅱ microsatellite instability on diagnosing the early prostate cancer. Method Fluorescent multiplex PCR was used to amplify annexin Ⅱ three loci in 16 prostate cancer tissues,16 benign prostatic hyperplasia tissues and 16 normal blood samples.Then,the 48 PCR products were collected and the differences and relationship between them were analyzed. Result Annexin Ⅱ D15S1185,D15S1292 loci have the same size PCR product in all the samples;AN2-di-repeat-NH dinucleotide repeats locus performs differences in size of 134 bp.Significant differences exist between prostate cancer tissues and normal blood samples(χ2=9.087,DF=1,P=0.003).Prostate cancer tissues and benign prostatic hyperplasia tissues didn't have obviously significant difference.(χ2=1.997,DF=1,P=0.158).Benign prostatic hyperplasia tissues and normal blood samples have significant trend (χ2=3.405,df=1,P=0.065). Conclusion The polymorphisms of a novel AN2-di-repeat-NH dinucleotide repeats locus may be a warning indicator of early prostate cancer.
