罗格列酮对糖尿病大鼠肾脏Smad7、TGF-β_1及Ⅳ型胶原表达的影响及意义

摘  要:目的 观察罗格列酮对链脲佐菌素诱导的实验性糖尿病大鼠肾脏的保护作用及机制.方法 选择鼠龄3个月左右SD雄性大鼠,体质量210 ~250 g,以60 mg/kg体质量的链脲佐菌素腹腔注射制备糖尿病大鼠模型,选择制模成功大鼠20只进行分组实验:罗格列酮给药组10只(E组)、生理盐水对照组10只(C组).E组以1 mg/kg体质量罗格列酮灌胃,C组则以相应剂量生理盐水灌胃,1次/d,连续4周,期满后摘取肾脏标本以4%磷酸盐缓冲液甲醛固定备检.采用免疫组织化学观察肾脏Smad7、转换生长因子β1(TGF-β1)及Ⅳ型胶原表达水平,免疫酶联吸附(ELISA)检测GHbA1c.结果 E组和C组肾脏组织Smad7 、TGF-β1、Ⅳ型胶原表达阳性面积百分率对数(ln)分别为:(2.49±1.43)%、(2.17±1.43)%(P<0.01),(2.52±0.89)%、(2.66±0.13)%(P<0.01),(2.58±0.15)%、(2.77±0.14)%(P<0.01).E组和C组GHbA1c水平对数(ln)分别为(3.49±0.21)%、(3.48%±0.17)%(P>0.05).结论 罗格列酮具有通过调节实验性糖尿病大鼠肾脏Smad7 、TGF-β1表达,抑制基质纤维化,对糖尿病肾病发病具有保护显著保护作用,而且此种效果并非由于罗格列酮的降糖作用.ObjectiveTo study the roles and protective mechanisms of rosiglitazone on nephropathy of streptozotocin-induced diabetic rats.MethodsSelecting age of 3 months,body weights 210-250 g male SD rats to make diabetic rat models induced by streptozotocin,selected 20 successful diabetic model rats and divided them into two groups randomly:experimented group(group E,n=10) fed with 1mg/kg rosiglitazone dissolved with normal solution,and control group(group C,n=10) fed with normal saline solution every day respectively,continued for 4 weeks,and the kidney specimens were removed and fixed with 4% phosphate buffer formalin,then GHbA1c was detected with ELISA,Smad7,TGF-β1 and Ⅳ collagen in kidneys by immunohistochemistry.ResultsThe mean levels(ln) of GHbA1c in group E and group C were (3.49±0.21)% and (3.48±0.17)% respectively(P〉0.05).The positively expressed area percent rates(ln) of Smad7,TGF-β1 and Ⅳ collagen in group E and group C were (2.49±1.43)%,(2.17±1.43)%(P〈0.01);(2.52±0.89)%,(2.66%±0.13)% (P〈0.01); (2.58±0.15)%,(2.77%±0.14)%(P〈0.01) respectively.ConclusionRosiglitazone may play very important protecting roles in the pathogenesis of diabetic nephropathy in experimental diabetic rats by upregulating the expressions of Smad7,which inhibits the Ⅳ collagen expressions induced by TGF-β1,which are not correlated with rosiglitazone's lowering glucose effect.
