摘  要:目的 探讨脊髓背根神经节自噬与大鼠术后持续性疼痛形成的关系.方法 雄性成年SD大鼠,体重220~250 g,随机分为3组(n=12):空白对照组(C组)、假手术组(S组)、皮肤/肌肉切口牵拉痛(SMIR)术后持续性疼痛模型组(SMIR组).SMIR组皮肤/肌肉切口牵拉痛模型手术,S组行皮肤切开,但不进行组织牵拉,C组不进行手术操作.各组分别于术前1 d、术后1、3、7、12 d时,测定机械缩足反应阈值(MWT).上述各时点痛阈测试完毕后,术后第12天测试MWT结束后每组随机取4只大鼠,取手术侧L4~5背根神经节电镜下观察自噬体情况,每组随机取4只大鼠,取手术侧L4~6背根神经节,采用Western blot测定自噬标记蛋白LC3Ⅱ表达.结果 与S组比较,SMIR组术后3、7、12 d MWT降低(P〈0.05),手术侧脊髓背根神经节LC3Ⅱ表达上调(P〈0.05).电镜结果显示,与S组和C组相比,SMIR组手大鼠术侧背根神经节可见明显自噬结构.结论 脊髓背根神经节自噬可能参与了大鼠术后持续性疼痛的形成.Objective To investigate the correlation between autophagy in dorsal root ganglia and persistent post- operative pain. Methods Male Sprague - Dawley rats, weighting 220 - 250 g, were randomly divided into 3 groups (n = 12) : Naive control group (Group C), Sham control group (Group S) and SMIR operation group (Group SMIR). Pain behavior was assessed by mechanical paw withdrawal threshold (MWT) to von Frey filament stimulation 1 day before and 1, 3, 7, 12 days after operation. L4_5 dorsal root ganglia of SMIR rats were investigated with electro microscope. Ex- pression of autophagy maker LC3 11 was qualified by Western blotting. Results Compared with Group C and Group S, MWT was significantly reduced 3, 7, 12 days after SMIR surgery (P 〈 0. 05 ). LC3 11 in ipilateral side of dorsal root ganglia was up - regulated following SMIR surgery. The electron microcopy showed obvious autophagy structure in SMIR rats. Conclusion Autophagy in dorsal root ganglia may participates in the pathogenesis of persistent postoperative pain.
