摘  要:对 2 0个来源及抗性不同的海岛棉和陆地棉品种和 2个陆地棉 ( S) /海岛棉 ( R)杂交组合的亲本、F1、 F2 世代的 POX同工酶研究表明 ,接种黄萎病菌前后 ,棉花叶片中的 POX同工酶酶谱发生明显变化 ,抗病品种与感病品种的阴性 POX同工酶谱带均由原来的 3条增至 6条 ,但两者之间不存在明显差异 ;阳性 POX同工酶谱带接种前后均只有 1条 ( POX- PC1) ,抗病品种接种前后该酶带无明显变化 ,而感病品种接种后活性增强。遗传分析表明 ,海岛棉对黄萎病的抗性和 POX- PC1酶带基因具有显性单基因遗传特征 ,并推测 PC1基因受抗病基因调控 ,在生化水平上进一步证明海岛棉抗性为显性单基因遗传。POX在棉花抗黄萎病中的作用并非十分重要 ,但 PC1酶带的表现与棉株黄萎病抗性显著相关 ,PC1酶带弱 ,棉株抗病 ,PC1酶带强 ,棉株感病 ,从而证明 POX-Peroxidase(POX) isozyme patterns were investigated in the materials including P 1, P 2, F 1 and F 2 of two crosses between susceptible Upland cotton cultivars and a resistant Sea Island cotton variety, and 20 cottons belonging to G. barbadense and G. hirsutum with different levels of Verticillium wilt resistance. There were three POX isozyme bands of anode in both the susceptible and resistant cottons when no pathogen stress, and after inoculation, three new bands of anode appeared in all the materials. On the other hand, one band of cathode, named POX PC1, was different between the susceptible and the resistant materials before inoculation, and the band of the susceptible varieties was more intensive than that of the resistant. After inoculation, the PC1 band of the resistant was still weak, but the band of the susceptible was much stronger than that of uninoculated plants. Through the analyses to the PC1 band of F 1 and F 2 populations, it was shown that almost all the bands of F 1 plants were weak, and the segregation ratios of weak and strong bands in F 2 populations were 3∶1. Since the resistance of the G. barbadense variety was controlled by a single gene, the PC1 band gene with the characteristic of single gene could be regulated or controlled by the resistant gene. In the F 2 generation of two combinations, it was observed that there was a significant relationship between intensiveness of PC1 band and Verticillium wilt resistance, with the performance of light colored band plant showing resistant and deep colored band plants susceptible. Therefore, the POX PC1 isozyme can be used as a genetic marker applicable to the future cotton breeding.
