摘  要:为解析杜仲异戊烯基焦磷酸异构酶基因序列信息和深入研究基因功能,以杜仲叶片cDNA为模板,采用反转录RCR及RACE技术分离出杜仲IPI基因全长的cDNA克隆。测序及生物信息学分析结果得出基因序列全长1 231 bp,共编码306个氨基酸残基,细胞定位于叶绿体。推导蛋白相对分子质量34.65 kD,理论等电点(pI)5.40,为疏水混合型蛋白质,属Nudix水解酶蛋白家族,基序类型4种,含10个潜在功能位点,三级结构以单体形式存在,主要保守基序位于分子结构中部。系统进化分析推测杜仲IPI蛋白与欧洲榛IPI蛋白亲缘关系最为接近。In order to analyze sequence information and further research the function of isopentenyl pyrophosphate isomerase gene in Eucommia ulmoides, taking eDNA in E. ulmoides leaf as template, IPI gene eDNA was isolated from leaves ofE. ulmoides by the method of reverse transcription polymerase chain reaction (RT-PCR) and rapid amplification of cDNA ends (RACE) technique. The results of sequencing and bioinformatics analysis show that the sequence of EuIPI gene is 1 231 bp and encode 306 amino-acid residues, and the cellular localization of the gene is in the chloroplasts. The relative molecular mass of deduced EuIPI protein is 34.65 kD and the theoretical isoelectric point is 5.40, and the protein is of an hydrophobic hybrid protein and belongs to the Nudix hydrolase protein family. EulPI protein has four types of motifs and 10 potential functional sites, the tertiary structure of the deduced protein exists by the monomer form, and the main conservative motif is located in the central of molecular structure of the protein. The results of phylogenetic analysis indicate that the evolutionary relationship of EuIPI protein is closest to IPI protein in Corylus avellana.
