摘 要:目的探讨广东汉族人群中脂联素基因(ADIPOQ)的单核苷酸多态性(SNP)与2型糖尿病的关联。方法采用SNPscanTM多重SNP分型试剂盒,对筛选的2个tagSNP(rs822396、rs3774261)在595名2型糖尿病患者和725名健康对照中进行基因分型,并应用χ2检验、Logistic回归模型等方法分析SNP位点与2型糖尿病的关联性。结果调整性别、BMI和甘油三酯等因素后,rs822396在等位基因关联分析和基因型关联分析中均无统计学意义(P>0.05),rs3774261在等位基因关联分析中亦无统计学意义[P=0.077,OR(95%CI)=1.18(0.98,1.41)],但在显性模型下的基因型关联分析中有统计学意义[P=0.028,OR(95%CI)=1.36(1.03,1.79)]。结论在广东汉族人群中,rs822396与2型糖尿病无关联,rs3774261与2型糖尿病的遗传易感性有关,但其具体机制尚需进一步探讨。Objective To evaluate the association between two single nucleotide polymorphisms located in ADIPOQ gene and type 2 diabetes mellitus in Han population in Guangdong Province. Methods Two tagSNPs (rs822396 and rs3774261) were picked out from ADIPOQ gene. Then SNP genotyping was performed using SNPscanTM Kit in two cohorts, including a total of 595 cases and 725 controls. Finally, Chi-square test, logistic regression model and other methods were employed in the following statistical analysis. Results No association between SNP rs822396 with T2DM was found in allelic association analysis and genotypic association analysis (P〉0.05) after adjustment for sex, BMI and TG, rs3774261 also had no association in allelic gene association analysis[P=-0.077, OR (95% CI) =1.18 (0.98, 1.41) ]. However, the GG+GA genotype of SNP rs3774261 was associated with T2DM in dominant model [P=-0.028, OR (95% C/) =1.36 (1.03, 1.79) ]. Conclusion SNP rs822396 of ADIPOQ gene is not associated with T2DM. However, rs3774261 is associated with increased risk of T2DM in Han population in Guangdong Province, and additional studies are needed to delineate further the role of this common variant in the pathogenesis of T2DM.
