摘 要:评价广东省肇庆地区地方柑橘品种与其他部分柑橘属和近缘属植物的亲缘关系,为其分类和育种提供理论依据。从34对柑橘SSR引物中筛选到13对多态性较好的SSR引物,对8份肇庆地方柑橘品种和22份柑橘属、枳属、酒饼簕属植物材料进行PCR扩增,并采用1.5%琼脂糖凝胶电泳检测,用遗传相似性和聚类分析等方法分析检测结果。13对引物共扩增出83条谱带,多态检出率为100%,通过聚类分析能够有效的区分柑橘属、枳属和酒饼簕属,以及柑橘属内的种、品种甚至品系。结果表明SSR分子标记技术能够用于肇庆柑橘种质资源分类地位研究,并且供试的肇庆地方柑橘品种亲缘关系较近。The results of present study in the evaluation of genetic relationship between Zhaoqing local citrus germplasm and other citrus cuhivars together with related genera by simple sequence repeat (SSR) molecular marker technique had provided solid supporting evidences for their classification and breeding. This thesis selected 13 pairs of SSR primers with better polymorphism from 34 pairs of SSR citrus primers and carried out PCR amplification in 8 samples of Zhaoqing local citrus cultivars, 20 samples of other citrus cultivars, 1 sample of Poncirus and 1 sample of Atalantia (used as out- goup), with the amplification product tested in 1 x TBE buffer solution using 1.5% agarose gel electrophoresis. Cluster analysis was performed using UPGMA, and a cluster tree was built using the NTSYSpc2.1 analysis software, with the genetic similarity coefficient calculated. A total of 13 primer pairs with high polymorphism were selected from the 34 pairs of SSR. The codes of primer sequence with polymorphism were TAA3, CTT01, TAA15, SS18, TAA1, SS16, CMS30, CAC19, CMS24, CSSR036, CMS14, CMS21, CMS20. A total of 83 polymorphic bands were amplified. Each primer had an average of 6.4 polymorphic bands. The detection rate of polymorphism was 100%. Clustering analysis can reveal genetic diversity and differentiate citrus, Poncitrus, and Atalantia genera from one another and identify species, breeds, and strains of citrus. It technique can be applied in the classification study of had been proven effective that SSR molecular marker Zhaoqing local citrus germplasm resources. And it also showed that the tested materials had a closer genetic relationship.
