摘  要:为了实现对一种农用抗生素的提取及柱层析分离,采用易于蒸发浓缩的丙酮替代常用的甲醇作提取剂,加速了有效成分的分离纯化进程,并有助于保持有效成分的生物活性;采用3因素、3水平正交试验设计法,探寻了该农用抗生素粗提物的浸提条件;借助极差分析法和方差分析法,最终确定最佳浸提条件为:100%丙酮、3倍物料体积的浸提液、浸提时间为2 h。采用硅胶柱层析法对浸提出的有效成分进行了分离纯化,以乙酸乙酯作为洗脱液,通过1次过柱分离除去了大量杂质,有效成分的回收率可达到83%以上,为有效成分的进一步分离纯化奠定了基础。In order to extract and isolate an agro-antibiotic from the fermentation products from a soil rare actinomycetes strain, acetone was chosen as extracting agent instead of more commonly used methanol as extracting agent and ethyl acetate as column chromatography eluent. A 3 levels and 3 factors orthogonal test design method was used to optimize extraction technology. From the data processing results by extremum difference analysis and variance analysis method, the optimal extraction condition was to extract with 3 fold acetone with 2 hours. Silica gel column chromatography was used to separate the active ingredients and ethyl acetate as eluent. A large amount of impurities was removed by once column chromatography and the recovery was more than 83%, which built foundation for further separation and purification of effective components.
