摘  要:目的观察冠心病患者脂联素(APN)-11377C>G基因多态性与阿托伐他汀调脂作用的关系。方法用自身对照方法,126例冠心病患者分为冠心病血脂异常组(CHD1)和正常组(CHD2组),阿托伐他汀每日20 mg,共观察12周;用聚合酶链反应-限制性片段长度多态性法,对其脂联素基因-11377C>G位点单核苷酸多态性进行基因型分析;研究脂联素基因多态性与阿托伐他汀疗效间的关系。结果脂联素基因-11377C>G位点的基因型为GG、CG及CC型;G等位基因频率和GG型频率:CHD1组均明显高于CHD2组(P<0.05);在阿托伐他汀干预第4,12周,TC、TG及LDL-C在各基因型较治疗前均明显下降,以第4周疗效显著;调脂疗效各基因型差异无统计学意义。APN升高在CC基因型较CG+GG组明显(P<0.05)。结论脂联素基因多态性与阿托伐他汀疗效无明显相关性。Objective To investigate the association between the adiponectin (APN)-11377C〉G gene polymorphism and the effects of atorvastatin. Methods Blood samples were collected from 126 patients with coronary heart disease (CHD). The patients were divided into dyslipidemia group (CHD1) and normal blood lipids group (CHD2). Polymorphism of adiponectin-11377C〉G was detected by the method of polymerase chain reaction fragment length polymorphism.The relationship between was investigated. Results The adiponectin gene-11377C〉G sites existing gene polymorphism were compared between patients with the 3 genotypes being GG, CG and CC. The GG genotype and G allele gene frequency of CHD1 group was significantly higher than the CHD2 group(P〈0.05). The 4th and 12th week of atorvastatin therapy, TC and TG and LDL-C significantly decreased, and lipid-efficacy of the 4th week was more significant. There was no statistical difference on lipid-efficacy of atorvastatin between each genotype of the APN-11377C〉G. The APN concentration of CC group was significantly higher than that of CG+GG group(P〈0.05).Conclusion There is no statistical relationship between the APN-11377C〉G gene polymorphism and atorvastatin lipid-efficacy in CHD patients.
