摘 要:Gbve1是海岛棉中的1个抗棉花黄萎菌基因,前期研究表明它能介导棉花与拟南芥对落叶型与非落叶型黄萎菌的抗性。为了揭示该基因的作用机制,对其细胞定位进行了研究。结果表明:该基因定位在植物细胞膜上;并检测了胼胝质沉积和活性氧积累情况,发现转基因拟南芥在黄萎菌侵染后胼胝质沉积,过氧化氢含量明显高于对照植株。推测:Gbve1是1个膜定位蛋白基因,它能响应黄萎菌的侵染,诱导拟南芥产生抗病性。Gbvel ,which was isolated from the island cotton cuhivar,encodes a cell membrane receptor-like protein against Verticillium dahliae. To further reveal the mechanism of action of Gbvel gene ,Agrobacterium-mediated transient expression technology was used to study its cellular localization in tobacco. The results showed that the protein was located in the plant cell membrane. In order to study the mechanism of resistance, we examined callose deposition in Gbvel-transgenie Arabidopsis post E dahliae infection. The results showed the callose deposition was significantly stronger than that of the control plants ;reactive oxygen test results showed that hydrogen peroxide in transgenic Arabidopsis after inoculation was also significantly higher than that in the control plants. Collectively, we gained a better understanding of the resistance mechanism of Gbvel gene, and provided a scientific basis for its effective use in agriculture.
