摘  要:目的分析抗炎抗凝双效融合蛋白TAP-SSL5对动物的药理作用,为其安全性用药提供实验依据。方法取昆明(KM)小鼠,经尾静脉分别注射1、3和9 mg/kg TAP-SSL5,观察其对小鼠神经系统的影响;取SD大鼠,经尾静脉分别注射1和3 mg/kg TAP-SSL5,设溶剂[20 mmol/L甘氨酸(Gly)-NaOH缓冲液,pH 10.6]为对照组,观察其对呼吸系统和心血管系统的影响。结果 TAP-SSL5对KM小鼠一般行为状态、协调机能和戊巴比妥钠阈下剂量催眠的协同作用无明显影响;对SD大鼠的血压、呼吸频率和心率无明显影响,与溶剂对照组比较,差异无统计学意义(P均>0.05)。结论融合蛋白TAP-SSL5对动物的神经系统、呼吸系统和心血管系统无明显影响,表明其具有较好的安全性,且无明显的毒副作用。Objective To analyze the general pharmacology of anti-inflammatory and anticoagulant tick anticoagulant peptide ( TAP )-staphylococcal superantigen like protein-5 (SSL5) fusion protein in animals and provide an ex- perimental basis for safe administration of the drug. Methods KM mice were injected i.v. with TAP-SSL5 at dosages of 1, 3 and 9 mg/kg respectively, and observed for effect on nerve system. SD rats were injected i.v. with TAP-SSL5 at dosages of 1 and 3 mg/kg respectively, using the solvent (20 mmol/L Gly-NaOH buffer, pH 10. 6) as control, and observed for effect on respiratory and cardiovascular systems. Results TAP-SSL5 showed no significant effect on the general status, coordination function and the synergistic action of pentobarbital sodium at subthreshold dosage of KM mice. However, the blood pressure, respiratory frequency and heart rate of SD rats treated with TAP-SSL5 showed no significant differences with those in control group (P 〉 0. 05 ). Conclusion TAP-SSL5 showed no significant effect on nerve, respiratory and cardiovascular systems of animals, indicating good safety and no significant toxicity of the fusion protein.
