摘 要:无菌采集陕北白绒山羊瘤胃内容物,梯度稀释后涂布CaCO3-MRS培养基,厌氧培养分离乳酸杆菌,并对分离菌株的生长、发酵液pH、酸耐受性、胆盐耐受性、抑菌性及有无内源质粒等生物学特性进行了比较研究。结果显示:从陕北白绒山羊瘤胃液中共分离出11株乳酸杆菌,经16S rDNA比对鉴定,分别是植物乳酸杆菌(Lactobacillus plantarum)4株(ZW-4、ZW-8、ZW-10、ZW-11),干酪乳酸杆菌(Lactobacillus casei)2株(GL-1、GL-5),罗伊乳酸杆菌(Lactobacillus reuteri)2株(LY-6、LY-7),短乳杆菌(Lactobacillus brevis)1株(DR-9),棒状乳酸杆菌(Lactobacillus coryniformis)1株(BZ-2)及弯曲乳酸杆菌(Lactobacillus curvatus)1株(WQ-3)。在相同培养条件下,植物乳酸杆菌ZW-4和干酪乳酸杆菌GL-1生长、产酸以及抑菌能力较强,同时植物乳酸杆菌ZW-4对胆盐和酸的耐受能力强于其他菌株;在上述菌株中,仅罗伊乳酸杆菌LY-7没检测到内源质粒,而植物乳酸杆菌ZW-4有6个内源质粒,明显多于其他几株乳酸杆菌。结果表明,分离自陕北白绒山羊瘤胃的植物乳酸杆菌ZW-4和干酪乳酸杆菌GL-1具有优良生物学特性,初步具备了作为益生菌使用的基本条件。Rumen contents were collected under gnotobasis from Shanbei White Cashmere Goat, and then Lactobacillus was isolated by ser ial dilution and anaerobic incubation in MRS-CaCO3 culture media. The growth, fermentation broth pH, acid resistance, bile salt tolerance, antibacterial activity and endogenous plasmids of Lactobacillus were analyzed. Eleven strains of LactobaciUi were screened from Shanbei White Cashmere Goat rumen. According to the 16S rDNA identification, 4 strains were L. plantarum (ZW-4, ZW-8, ZW- 10, ZW- 11 ) , 2 strains were L. casei ( GL- 1, GL-5 ), 2 strains were L. reuteri ( LY-6, LY-7) , 1 strain was L. brevis ( DR-9), 1 strain was L. coryniformis (BZ-2) and 1 strain was L. curvatus (WQ-3). In the same culture condition, L. plantarum ZW-4 and L. casei GL- 1 showed better growth, higher acid-production efficiency and antibacterial abilities. Moreover, L. plantarum ZW-4 was more tolerant than the other strains under bile salts and acid. L. plantarum ZW-4 had 6 endogenous plasmids, which was the most in all the Lactobacilli isolates. However, no endogenous plasmids were found in L. reuteri LY-7. We found that L. plantarum ZW-4 and L. casei GL-1 from Shanbei White Cashmere Goat rumen had excellent biological characteristics, and met the general requirement of probiotics.
