摘  要:目的考察低剂量丹墨胶囊对11β-羟基类固醇脱氢酶(11β-HSD)基因表达的影响,分析丹墨胶囊与糖皮质激素相互作用的具体机制。方法雄性SD大鼠连续14 d灌胃给予丹墨胶囊0.216 g/kg后,定量RT-PCR法测定大鼠肝、肾、胸腺、脾、脂肪、肌肉、心脏、肺、小肠、睾丸等组织11β-HSD1、11β-HSD2基因表达。结果丹墨胶囊显著诱导肝脏组织11β-HSD1基因表达,显著抑制大鼠心脏、肺、脾、肌肉、胸腺组织11β-HSD1的基因表达,但对肾脏、脂肪、睾丸、脑、小肠组织的11β-HSD1基因表达无明显影响;抑制心脏、脾、肌肉组织11β-HSD2基因表达,对肝脏、肾、脂肪、睾丸、肺、脑、小肠和胸腺组织的11β-HSD2基因表达无明显影响。结论丹墨胶囊可诱导大鼠肝脏组织中11β-HSD1基因表达,影响糖皮质激素体内活化。Objective To investigate effect of low dose of Danmo capsules on gene expression of 11 β-hydroxysteroid dehydrogenase (11β-HSD), then to analyze the specific mechanism of interaction between Danmo capsules and glucocorticoids. Methods After oral administration of Danmo capsules (0. 216 g/kg) in male SD rats for 14 consecutive days, gene expression of 11β-HSD1 and 11β-HSD2 in tissues of liver, kidney, thymus, spleen, fat, muscle, heart, lungs, small intestine, testis tissues were determined with quantitative RT-PCR method. Results Danmo capsules significantly induced gene expression of 11β-HSD1 in liver, but inhibited gene expression of 11β-HSD1 in heart, lung, spleen, muscle and thymus. Danmo capsules had no significant effect on gene expression of 11β-HSD1 in kidney, fat, testis, brain and small intestine. Danmo capsules significantly inhibited gene expression of 11β-HSD2 in heart, spleen and muscles. However, there was no significant effect on gene expression of 11β-HSD2 in liver, kidney, fat, testis, lung, brain, small intestine and thymus. Conclusions Danmo capsules could induce activity of corresponding enzyme through affecting gene expression of 11β-HSD1 in liver tissues, then affect in-vivo activation of glucocorticoids.
