摘  要:目的研究慢性丙型肝炎患者外周血中CD4+Foxp3+调节性T细胞(regulatory T cell,Treg)的变化及其临床意义。方法分别采用流式细胞术、实时定量逆转录聚合酶链反应(reverse transcription polymerase chain reaction,RT-PCR)和酶联免疫吸附法(enzyme linked immunosorbent assay,ELISA)分别检测60例慢性丙型肝炎患者及40名健康对照者外周血CD4+Foxp3+Treg细胞百分率、核转录因子foxhead winged-helix box protein 3(Foxp3)mRNA的水平以及血浆转化生长因子-β1(transforming growth factor-β1,TGF-β1)的浓度。结果慢性丙型肝炎患者外周血中CD4+Foxp3+Treg细胞百分率、Foxp3mRNA表达水平及血浆TGF-β1浓度与正常对照组相比均明显升高,差异有统计学意义(P<0.05)。结论慢性丙型肝炎患者外周血中CD4+Foxp3+Treg细胞及TGF-β1水平增高,Treg细胞可能参与了慢性丙型肝炎发病过程。Objective To explore the changes of CD4 + Foxp3 + Treg cells in the peripheral blood of patients with chronic hepatitis C and their clinical significance. Methods The percentage of CD4 + Foxp3 + Treg cells was assessed by flow cytometry and the mRNA level of Foxp3 was detected by real-time quantitative reverse transcription polymerase chain reaction (RT-PCR) in 60 patients with chronic hepatitis C and 40 healthy controls. The plasma levels of TGF-β1 were measured by enzyme linked immunosorbent assay (ELISA). Results The percentage ofCD4 + Foxp3 + Treg cells,the mRNA expression of Foxp3 and the plasma level of TGF-β1 were significantly higher in patients with chronic hepatitis C than those in healthy controls (P 〈 0. 05). Conclusion The results suggest that Treg cells may be associated with the development of chronic hepatitis C.
