摘 要:目的探讨高速逆流色谱(HSCCC)法提纯黄柏与五倍子配伍抗菌活性物质对粪肠球菌体外抑茵效果的影响。方法选择150株临床分离野生粪肠球菌株,随机分为Ⅰ组、Ⅱ组与Ⅲ组,每组各50株,均配制成2000cfu/ml^3000cfu/ml的菌液。黄柏与五倍子配伍药做成中药煎剂,配制成浓度成1g/ml为A药液,高速逆流色谱法提纯黄柏与五倍子配伍抗菌活性物质,配制成浓度成1g/ml为B药液;Ⅰ组将粪肠球菌液涂到用A药液与M-H培养基混合的培养基上,Ⅱ组将粪肠球菌液涂到用B药液与M-H培养基混合的药液培养基上,Ⅲ组将粪肠球菌液涂到M-H培养基上,上述培养基均贴上环丙沙星(CIP)、红霉素(ERY)、四环素(TE)、青霉素(PEN)、氯林可霉素(ClE)、苯唑青霉素(OXA)和克拉霉素(CLA)药敏纸,并观察培养基药敏情况。结果Ⅰ组的CIP、ERY、TE、PEN、ClE、OXA和CLA耐药率分别为26%、40%、36%、30%、42%、38%和24%,Ⅱ组Ⅲ组为10%、26%、14%、8%、18%、24%、4%,和76%、92%、100%、88%、96%、90%、60%,对CIP、ERY、TE、PEN、ClE、OXA和CLA耐药率Ⅲ>Ⅰ组>Ⅱ组,三组之间比较,差异均有统计学意义(P<0.05)。结论高速逆流色谱法提纯黄柏与五倍子配伍抗菌活性物质对临床分离的粪肠球菌菌株具有较强的抑菌活性,其抑茵作用比黄柏与五倍子配伍药做成中药煎剂强。Objective To explore the influence of bacteriostasis on Enterococcus faecalis by antibacterial substance purified from combination of Cortex Phellodendri and Galla Chinensis by high- speed counter- current chromatography.Methods 150 strains of clinical wild Enterococcus faecalis isolates were selected and randomly divided into Group I, Group II and Group III, 50 strains in each group were made up as 2 000 cfu/mL^3 000 cfu/mL Enterococcus faecalis solution. The combination of Cortex Phellodendri and Galla Chinensis were formulated as solution A of 1 g/mL. Antibacterial substance purified by HSCCC was prepared as solution B with the concentration of 1g/mL. The Enterococcus faecalis solution was smeared on mixed culture medium of M-H medium and solution A in Group I, mixed cultured medium of M-H and solution B in Group II, and merely on M-H in Group III. The above- mentioned culture medium were all pasted with the drug sensitivity paper of ciprofloxacin(CIP), erythrocin(ERY), tetracycline(TE), penicillin(PEN),clindamycin(ClE) and oxacilin(OXA) and clarithmmy cinl(CLA) so as to observe the drug sensitivity of the culture medium. Results The resistance rates of CIP, ERY,TE, PEN, ClE, OXA and CLA in Group I were 26%,40%,36%,30%,42%,38% and 24%, respectively;and those in Group II were 10%,26%,14%,8%,18%,24% and 4%, respectively and in Group III as 76%,92%,100%,88%,96%,90% and 60%.The resistance rates of CIP, ERY, TE, PEN, ClE, OXA and CLA in Group IIIGroup I Group II,and the statistically significant differences of drug resistance rates among the three groups were found(P 0.05).Conclusions The antibacterial substance purified from the combination of Cortex Phellodendri and Galla Chinensis purified by HSCCC has a strong antibacterial activity on clinical Enterococcus faecalis isolates,and its antibacterial action is stronger than the decoction of combination golden cypress and Chinese Gall.
