摘 要:目的:选取低氧反应基因——诱导型一氧化氮合酶(inducible nitric oxide synthase,iNOS)基因序列上rs10459953、rs1060822单核苷酸多态性(single nucleotide polymorphism,SNP)位点,研究其与低氧训练后生理表型指标关联性。方法:采用关联研究方法(AssociationStudy),聚合酶链反应-限制片段长度多态性(PCR-RFLP)实验解析技术,选取35名健康受试者在模拟海拔2500 m高度进行4周高住-高练-低训(living high-exercise high-training low,Hi-Hi-Lo)。研究iNOS基因SNP rs10459953和SNPrs1060822与低氧训练后最大摄氧量(maximal oxygen uptake,VO2max)、血红蛋白(hemoglobin,Hb)、红细胞数(red blood cell,RBC)等生理表型指标关联性。结果:1)4周低氧训练后,SNP rs10459953不同基因型之间VO2max、Hb、RBC变化量均无显著性差异。2)4周低氧训练后,SNP rs1060822不同基因型间Hb、RBC变化量有统计学意义(P<0.05),其中CT基因型受试者Hb、RBC显著增加(P<0.05),且在低氧环境下定量负荷实验中,SNP rs1060822的CT基因型受试者低氧训练后期SpO2动态曲线显著高于初期(P<0.05)。结论:iNOS基因SNP rs1060822与低氧训练后血象指标存在一定关联性,CT基因型表现出一定优势。Objective: In this study, we selected two single nucleotide polymorphisms (SNP), rs10459953 and rs1060822, in the hypoxia response gene, inducible nitric oxide synthase(iNOS) , to investigate association between these polymorphisms of iNOS gene and physiological phenotype parameters response to hypoxia training. Methods :35 healthy subjects completed a g weeks"living high - exercise high - training low" in a simulated ahitude of 2500m height, and using association - study of polymerase chain reaction- restriction fragment length polymorphism( PCR- RFLP) for genotype analysis. To investigate association between SNP rs10459953 and rs1060822 of iNOS gene and physi- ological phenotype parameters including maximal oxygen uptake (VO2max), hemoglobin (Hb), red blood cell ( RBC ) and other parameters after hypoxia training. Results : ( 1 ) After the 4 weeks hypoxia ' training,there was no significant difference in VOEmax, Hb or RBC between different genotypes of SNP rs10459953. (2) After the 4 weeks hypoxia training, carriers with CT genotype of SNP rs1060822 showed significantly higher increase in Hb and RBC ( P 〈 0.05 ). There were significant differences in carriers with CT genetype of SNP rs1060822 in SpO2 during period of exercising before and after 4 weeks hypoxia training ( P 〈 0.05 ) , and SpO2 curve was higher after hypoxia training. Conclusion :These findings suggested that SNP rM060822 of iNOS gene was possibly associated with hemogram parameters after hypoxia training, and CT genotype showed a certain advantage.
