中药清热利湿饮对HaCaT细胞表达IL-6mRNA、IL-8mRNA、IL-10mRNA及分泌IL-8、IL-10的影响

摘　　要：目的:揭示中药清热利湿饮对人永生化角质形成细胞HaCaT细胞表达IL-6mRNA、IL-8mRNA、IL-10mRNA和分泌IL-8、IL-10的作用机制。方法:以HaCaT细胞为研究对象,分别采用RT-PCR技术及ELISA方法观察中药清热利湿饮提取液对经TNF-α诱导活化的HaCaT细胞表达IL-6mRNA、IL-8mRNA、IL-10mRNA和分泌IL-8、IL-10的影响。结果:清热利湿饮对经TNF-α诱导活化的HaCaT细胞IL-6mRNA和IL-8mRNA的表达有下调作用,在药物浓度为10g/L和12.5g/L时,HaCaT细胞IL-10mRNA的表达量增强较为明显,与TNF-α诱导活化组相比,有显著性差异(P<0.05或P<0.01)。清热利湿饮对经TNF-α诱导活化的HaCaT细胞IL-8的分泌有不同程度的抑制作用,但对IL-10的影响不明显。结论:清热利湿饮可下调IL-6mRNA、IL-8mRNA的表达,上调IL-10mRNA的表达,且此作用随药物浓度的升高而增强;同时可降低细胞IL-8的分泌,但对IL-10的分泌无明显的作用。Objective：To reveal the mechanism of traditional Chinese medicinal Qingre Lishi Yin on the expressions of IL - 6mRNA,IL- 8mRNA,IL- 10mRNA and secretion of IL- 8, IL- 10 in HaCat cells. Methods：Using HaCaT cell as the research object, respectively to observe the effect of Qingre Lishi Yin extraction of IL -6mRNA, IL -8mRNA, IL - 10mRNA and the secre- tion of IL - 8, IL - 10 solution on the expression of activation HaCaT cells induced by TNF - a through RT - PCR and ELISA. Re- sults ： Qingre Lishi Yin could downregulate the expressions of IL - 6mRNA and IL - 8mRNA of HaCaT ceils induced by TNF-a. In the drug concentration for 10g/L and 12.5g/L, the expression of IL - 10mRNA increased significantly, and activated by TNF-a group, there was significant difference（ P 〈 O. 05 and P 〈 0.01 ）. Qingre Lishi Yin on HaCaT cell induced by TNF-a IL-8 activa- tion had different degrees of inhibition of secretion, but no obvious effect on IL - 10. Conclusion：Qingre Lishi Yin can reduce the expressions of IL - 6mRNA,IL - 8mRNA,up regulate the expression of IL - 10mRNA, and this effect increases with the concen- tration increasing;at the same time it can reduce the secretion of IL -8 cells,but no obvious effect on IL - 10 secretion.
