摘  要:【目的】观察滋肾育胎丸对900 MHz拟手机辐射雌性大鼠卵巢功能和卵巢ATM蛋白表达的影响。【方法】选用30只SD雌性大鼠随机分为正常组、辐射组、滋肾育胎丸组,正常组不接受辐射,辐射组和滋肾育胎丸组每天暴露于自制900 MHz手机辐射源4 h,连续30 d,滋肾育胎丸组每天灌胃滋肾育胎丸悬液,另外2组灌胃等容积生理盐水,阴道涂片选动情期处死大鼠。采用放射免疫法检测各组大鼠血清睾酮(T)、雌二醇(E2)含量,苏木精—伊红(HE)染色观察卵巢组织形态学变化,免疫组织化学法检测大鼠卵巢ATM蛋白表达。【结果】与正常组比较,辐射组大鼠血清E2水平显著降低(P<0.05),血清T含量无显著变化(P>0.05);辐射组卵巢颗粒细胞层数减少(P<0.05),颗粒细胞核ATM蛋白表达减弱;与辐射组大鼠比较,滋肾育胎丸组大鼠血清E2水平显著升高,卵巢颗粒细胞层数增多,ATM蛋白表达显著增强(均P<0.05)。【结论】拟手机辐射对大鼠卵巢功能有不良影响,可能与影响卵巢ATM蛋白表达有关;滋肾育胎丸对手机辐射大鼠卵巢损伤具有保护作用。Objective To study the impacts of Zishen Yutai Pills on the ovarian function and the expression of ataxia-telangiectasia mutated (ATM) gene protein in the female rats exposed to 900 MHz of mimic cellular phone radiation. Methods Thirty female SD rats were randomly divided into normal group, radiation group and Zishen Yutai Pills group. The rats in the normal group were free from radiation. The rats in the radiation group and Zishen Yutai Pills group were exposed to 900 MHz radiation from hand-made mimic cellular phones for 4 hours every day, lasting for 30 days. Meanwhile, the rats in Zishen Yutai Pills group were given intragastric administration of suspension of Zishen Yutai Pills every day, and the rats in other two groups were given oral administration of the same volume of normal saline. Rats were killed when they were at the estrum confirmed by vaginal smears. Serum testosterone (T) and estradiol (E2) contents in rats were detected by radioimmunoassay. The ovary histological and morphologic features were observed by HE staining, and the expression of ATM protein in the ovary was detected by immunohistochemical method. Results Compared with the normal group, the level of serum E2 was obviously decreased (P〈0.05), T content had no obvious changes (P〉0.05), the layers of ovarian granular cells were decreased, and the expression of ATM gene protein was lowered (P〈 0.05) in the radiation group. Compared with the radiation group, the serum E2 level was significantly increased (P〈0.05), the layers of ovarian granular cells were increased, and the ATM gene protein expression was high (P〈0.05) in the Zishen Yutai Pills group. Conclusion The exposure of 900 MHz of mimic cellular phone radiation is harmful to the rat ovarian function, which may be related to the decrease of ATM gene protein expression. Zishen Yutai Pills can protect the ovarian function from cellular phone radiation.
