摘 要:【目的】观察橘荔散结丸对子宫肌瘤模型大鼠雌激素受体(ER)、孕激素受体(PR)、表皮生长因子受体(EGFR)表达及组织病理学的影响,探讨橘荔散结丸治疗子宫肌瘤的作用机制。【方法】选用SD大鼠,随机分为空白对照组,模型对照组,西药对照组(米非司酮,剂量为4.7 mg/kg),中药对照组(桂枝茯苓胶囊,剂量为0.52 g/kg),橘荔散结丸低、中、高剂量组(中药低、中、高剂量组,剂量分别为9.86、19.73、39.45 g/kg);除空白对照组外,其他组均采用雌、孕激素负荷法复制子宫肌瘤大鼠模型,观察各组大鼠子宫肌层ER、PR和EGFR的表达及病理学改变。【结果】模型对照组大鼠子宫明显增大,子宫肌细胞排列紊乱,子宫肌层ER、PR和EGFR的表达均显著高于空白对照组(P<0.05);橘荔散结丸各剂量组子宫平滑肌排列趋于正常,子宫肌层ER、PR和EGFR的表达明显下降,其中中剂量组与模型对照组比较,差异有统计学意义(P<0.05)。【结论】橘荔散结丸治疗子宫肌瘤的作用机制可能与其能抑制子宫肌层ER、PR和EGFR的表达有关。Objective To investigate the effect of Juli Sanjie Pills (JSP) on the expression of estrogen receptor (ER), progesterone receptor (PR) and epidermal growth fhctor receptor (EGFR) in the myometrium of uterine leiomyomas rats, and to explore the therapeutic mechanism. Methods SD rats were randomized into blank control group, model group, western medicine control group (mifepristone in the dosage of 4.7 mg/kg) , Chinese medicine control group (Guizhi Fuling Capsules in the dosage of 0.52 g/kg) , and low-, middle- and high-dosage JSP groups (in the dosage of 9.86, 19.73, 39.45 g/kg, respectively) . Except for the blank control group, the rats in the other groups were given estrogen and progesterone load to induce uterine leiomyoma. After treatment, the pathological changes in the myometrium were observed, and the expression of ER, PR and EGFR in myometrial cells was detected by immunohistochemistry. Results The volume of uterus was enlarged, the myometrial cell arrangement was in disorder, and the expression of ER, PR and EGFR was increased obviously in the model group compared with those of the blank control group (P〈0.05) . In JSP groups, the uterine myometrial cell arrangement was in order, and the expression of ER, PR and EGFR in the uterine myometrial cells was decreased, the difference being significant between the middle-dosage JSP group and the model group (P〈0.05) . Conclusion The results suggest that the therapeutic mechanism of Juli Sanjie Pills for uterine leiomyoma is related with the inhibition of ER, PR and EGFR expression in the rat myometrial cells.
