摘 要:目的:探讨生大黄、清半夏、常山对正常大鼠胃肠功能和脑-肠轴β-内啡肽(β-EP)含量的影响,并分析与中药升降浮沉的相关性。方法:将64只Wistar大鼠分为两大组,每组随机分为4组,连续给药后,一组测定大鼠的结肠推进率和胃排空能力,另一组测定大鼠血浆、下丘脑、胃窦、小肠中β-内啡肽含量。结果:与对照组比较,生大黄能明显促进大鼠的胃肠蠕动功能,使大鼠下丘脑、胃窦β-EP含量降低明显(P<0.05),小肠中β-EP降低更明显(P<0.01),血浆中β-EP含量有上升的趋势;清半夏促进胃肠功能也比较明显,使大鼠胃窦、小肠中β-EP含量降低较明显(P<0.05),下丘脑和血浆中含量变化不大;常山可明显抑制大鼠的胃肠功能,使血浆中β-EP含量下降趋势明显(P<0.05),其他位点含量变化不明显(P>0.05)。结论:具有沉降趋势的生大黄与清半夏以及具有升浮药性的常山通过影响脑-肠轴中β-EP含量,改变胃肠的运动从而发挥其升浮或者沉降之药性。Objective:To investigate the effects of Crude Rhubarb,Rhizoma Pinelliae Preparata,and Dichroa febrifuga on gastrointestinal function and β-endorphin(β-EP) levels in brain-gut axis of normal rats and the correlation to upbearing,downbearing,floating and sinking theory. Methods:Sixty-four healthy Wistar rats were randomly divided into two groups. Each group was randomly divided into 4 sub-groups. After dosing,intesti- nal propulsion and gastric emptying tests were performed in one group. β-EP levels in the plasma,hypotha- lamus,gastric antrum and small intestine were measured in another group. Results:Compared with control group,the Crude Rhubarb group could significantly promot intestine emptying and decrease β-EP levels in the hypothalamus,gastric antrum and small intestine(P〈0.05 or P〈0.01),β-EP levels in plasma showed an increasing trend. The Rhizoma Pinelliae Preparata group had significantly promoted intestine emptying and decreased β-EP levels in the,gastric antrum and small intestine(P〈0.05),but with little changes in β-EP levels in the hypothalamusand plasma. The Dichroa Febrifuga group had a marked inhibitory effect on the digestive system and showed a significant declining trend in plasma β-EP level(P〈0.05),but no significant changes in β-EP levels were seen in other sites(P〉0.05). Conclusion:Dichroa Febrifuga shows upbearing and floating properties,while Crude Rhubarb and Rhizoma Pinelliae Preparata show downbearing and sinking properties,according to the relationships with β-EP levels in brain-Rut axis.
