摘  要:目的探讨花生四烯酸(AA)和二磷酸腺苷(ADP)浓度对血小板聚集的影响。方法随机收集47例门诊体检健康志愿者静脉血标本,枸橼酸钠抗凝,离心后分离富血小板血浆(PRP)和贫血小板血浆(PPP),AA(0.4,0.5,0.7,0.8,1.0,1.5和2.0mmol/L)或ADP(2.5,5,7.5,10,15,20,40和100μmol/L)各浓度上机,采用血浆比浊法测血小板聚集率。结果在诱导剂AA浓度在0.4~2.0mmol/L时,血小板聚集率为(53.9±29.5)%~(64.0±14.5)%(F=5.19,P〈0.05);在诱导剂ADP浓度在2.5~100/μmol/L时,血小板聚集率为(51.5±8.7)%~(74.9±7.1)0A(F=78.44,P〈0.05)。AA诱导血小板聚集在AA0.7mmol/L时血小板聚集率最高,随AA浓度进一步增高,聚集率有降低的趋势;ADP诱导血小板聚集率随ADP浓度逐渐增高而增高,ADP浓度与血小板聚集率呈线性相关(P〈0.05,r=0.7649)。结论诱导剂浓度对血小板聚集的影响明显,在PRP血小板浓度250×10-9/L左右时,诱导剂AA0.7mmol/L和ADP10μmol/L为诱导血小板聚集的适宜浓度。Objective To detect the effect of arachidonic acid (AA) and adenosine diphosphate (ADP) concentration on test results of platelet aggregation. Methods Blood samples of outpatient medical volunteers (n=47) were collected and antico- agulaged by sodium citrate,then platelet-rich plasma (PRP) and platelet-poor plasma (PPP) was obtained by centrifuging. The rate of platelet aggregation were detected following standard protocols by plasma turbidimetry with inducer of AA (0.4,0.5,0.7,0.8,1.0,1.5 and 2.0 mool/L) or ADP (2.5,5,7.5,10,15,20,40 and 100 μmol/L). Results When AA and ADP concentration was in the range of 0.4-2.0 mmol/L and 2.5-100 μmol/L respectively,the rates induced by AA and ADP were (53.9±29.5)%(64.0±14.5)% (F=5.19,P〈0.05) and (51.5± 8.7)%(74.9±7.1)% (F=78.44, P〈0.05) respectively. The rate of AA became maximal at the AA concentration of 0.7 mmol/L and then tended to decrease with a higher AA, the rate of ADP gradually increased with a higher concentration of ADP and showed a linear correlation (r =0. 764 9,P〈0.05). Conclusion The AA and ADP concentration of 0.7 mmol/L and 10 μmol/L seems to be the optimal concentration at the platelet concentration of 250× 109/L.
