摘  要:目的:研究紫草素对白介素(IL)-22刺激永生化角质形成细胞株HaCaT细胞增殖、迁移及分化的影响。方法:WST-1法检测紫草素对II-22刺激HaCaT细胞增殖的作用;Transwell法检测紫草素对IL-22刺激HaCaT细胞迁移的影响;Western blot法检测紫草素对IL-22刺激后HaCaT细胞分化相关蛋白兜甲蛋白(loricrin)表达的影响。结果:紫草素可抑制IL-22引起的HaCaT细胞增殖和移动;促进了IL-22刺激后HaCaT细胞分化相关蛋白的表达。结论:紫草素对IL-22介导的体外培养HaCaT细胞增殖及迁移有抑制作用并促进细胞分化。Objective: To investigate the effects of shikonin on the proliferation, migration and differentiation of HaCaT cells treated with interleukin (IL)-22. Methods: WST-1 and transwell assays were used to evaluate the inhibitory effects of shikonin on the proliferation and migration of HaCaT cells incubated with IL-22 respectively. The expression of loricrin(a marker protein of the differentiation of keratinocyte) on HaCaT cells was analyzed by Western blot. Results: The proliferation and migration of HaCaT cells induced by IL-22 were significantly inhibited by shikonin, the down-regulated expressions of loricrin on HaCaT cells induced by IL-22 were increased significantly by adding shikonin. Conclusion: In vitro, shikonin could inhibit the proliferation, migration and promote the differentiation of HaCaT cells mediated by IL-22.
