摘  要:目的百草枯是目前全球广泛使用的接触性除草剂,对人畜具有极强的肺毒性。文中旨在探讨水杨酸钠对百草枯急性中毒大鼠生存率及肺损伤的作用。方法取Wistar大鼠59只,按随机数字分组法分为正常对照组、水杨酸钠对照组、百草枯A组、百草枯B组、水杨酸钠A组、水杨酸钠B组。百草枯A组、水杨酸钠A组用百草枯(25mg/kg)腹腔注射染毒,百草枯B组、水杨酸钠B组用百草枯(35 mg/kg)腹腔注射染毒,水杨酸钠A组、水杨酸钠B组在染毒后2h分别腹腔注射水杨酸钠200 mg/kg、300 mg/kg干预;水杨酸钠对照组予腹腔注射水杨酸钠(300 mg/kg);正常对照组予腹腔注射等渗盐水。观察各组28 d生存率及肺病理学改变。结果肺病理学研究显示百草枯组第4天显示急性肺损伤,而正常对照组、水杨酸钠对照组肺组织未见明显变化。水杨酸钠治疗组的肺损伤病理学改变与百草枯组相比无明显改善。Masson染色显示百草枯染毒存活小鼠,不论有无水杨酸钠干预,后期均表现为纤维组织增生沉积、肺泡壁增厚。正常对照组和水杨酸钠对照组28 d生存率100%,百草枯A、B组28d生存率(36%、50%)显著低于正常对照组(100%),差异有统计学意义(P=0.035,P=0.050);与百草枯A组生存曲线比较,水杨酸钠A组的生存期无明显改善,差异无统计学意义(P=1.000),水杨酸钠B组与百草枯B组相比,生存期同样无明显改善,差异无统计学意义(P=1.000)。结论从28 d生存率和病理学角度观察,水杨酸钠对急性百草枯中毒大鼠的肺损伤无保护作用。Objective Paraquat, as a widely used contact herbicide, is a strong pneumotoxicant. This study is to investigate the effects of sodium salicylate (NASAL) on paraquat-exposed rats through observation of the 28-day survival rate and pathological changes in the lungs of the rats. Methods Fifty-nine Wistar rats were randomly divided into six groups: normal control, NASAL control, paraquat A, paraquat B, paraquat + NASAL A, and paraquat + NASAL B. The rats in the normal and NASAL control groups were intraperitoneally injected with saline and NASAL at 300 mg/kg, those in paraquat A and B with paraquat at 25 and 35 mg/kg, and those in paraquat + NASAL A and B with the same doses of paraquat followed 2 hours later by intraperitoneal injection of NASAL at 200 and 300 mg/kg, respectively. Then all the rats were observed for pathological changes in the lungs and the rate of survival for 28 days. Results Acute lung injury was observed at 4 days in the paraquat groups, but not in the normal and NASAL control groups. The pa-raquat + NASAL groups showed no significant improvement in lung injury as compared with the paraquat groups. Masson staining re-vealed diffuse thickness of the pulmonary alveolar wall with visible extensive fibrosis in the surviving rats of the paraquat groups regardless of NASAL intervention. The 28 day survival rate was 100% in the normal and NASAL control groups, significantly higher than in paraquat A (X^2 =6.491, P=0.011) and B (X^2 =4.500, P= 0. 034). The survival curves exhibited no significant differences be-tween the paraquat A and paraquat + NASAL A groups (X^2 = 0. 243,P = 0.622), nor between paraquat B and paraquat + NASAL B (X^2 = 0. 000, P = 0. 985 ). Conclusion Based on the observations of the 28 day survival rate and pathological findings, NASAL has no protective effect against paraquat poisoning-induced lung injury in rats.
