摘 要:总所周知,肿瘤坏死因子相关凋亡诱导配体(TNF-related apoptosis-inducing ligand, TRAIL)诱导多种肿瘤细胞及转化细胞发生凋亡,但对正常组织及细胞无损伤作用。近年研究发现,TRAIL信号通路不仅诱导细胞凋亡,而且具有促进增殖、介导耐药、诱导分化、促进转移以及增强侵袭等作用。TRAIL信号通路促细胞增殖作用对其临床应用于肿瘤治疗提出了挑战,但为血管和自身免疫性疾病的治疗提供新靶点。TNF-related apoptosis-inducing ligand (TRAIL) can induce the apoptosis of various types of tumor cells or transformed cells but sparing the normal tissues and cells. However, a num- ber of recent studies displayed that TRAIL could not only induce the apoptosis of cells, but also pro-mote the cell proliferation, mediate the drug resistance, induce the cell differentiation, promote the tumor metastasis and enhance the invasion of tumor cells. The promotion of cell proliferation by TRAIL signaling pathway poses a challenge to use of TRAIL as an anticancer agent in clinical appli-cation but nrovides novel therapeutic targets for vascular and autoimmune disorders.
