摘  要:建立反相高效液相色谱法测定甲氨蝶呤血药浓度的方法,以指导临床合理用药.采用Agilent ZOBAX SB-C18柱(150 mm×4.6 mm,5μm);流动相为0.025 mol/L磷酸二氢钠溶液(pH 6.30)-甲醇(76/24,V/V);流速为1.00 mL/min;检测波长为313 nm;柱温为35℃;1O%(V/V)高氯酸为蛋白沉淀剂.结果表明,血浆中甲氨蝶呤浓度在0.05~5.00 μg/mL(Y1=35.556 C+0.771,r=0.999 9)和5.00~100.00μg/mL(Y2=36.727C +1.700,r=0.9999)范围内与峰面积呈良好线性关系,平均回收率为99.03%和97.04%.日内精密度小于1%,日间精密度小于2.5%(n=6),定量浓度为0.05 μg/mL.该方法色谱条件稳定、操作简单、出峰时间快、线性范围宽、精密、准确、特异性强、回收率高,适用于甲氨蝶呤血药浓度的监测.A RP-HPLC method for detecting methotrexate (MTX) in human plasma was established.The sample was separated by Agilent ZOBAX SB-C18 (150 mm×4.6 mm,5 μm) column with mobile phase of sodium acetate buffer (pH 6.30)-methanol (76/24,V/V) at a flow rate of 1.0 mL/min.The wavelength of detection was 313 nm and the column temperature was 35 ℃.The plasma sample was treated with 10% perchloric acid for sedimentation.The results showed that the good linearity within 0.05~5.00 μg/mL(Y1=35.556 C+0.771,r=0.999 9)and 5.00~100.00 μg/mL(Y2 =36.727 C+1.700,r =0.999 9)were observed.The average recovery rates were 99.03% and 97.04%,respectively.The RSD with intra-day was less than 1% and the inter-day was less than 2.5%(n=6).The LOQ was 0.05 μμg/mL.The established method was simple,accurate,sensitive and reproducible with large-scale of detection range,suitable for monitoring the blood concentration of methotrexate.
