摘  要:目的对心肌梗死后心力衰竭大鼠经尾静脉注射腺病毒介导的β肾上腺素受体激酶抑制剂(βARKct),观察其对神经内分泌激素以及β肾上腺素受体信号传导的影响。方法体重250 g左右的雄性SD大鼠24只随机分为3组,其中2组结扎冠状动脉左前降支形成急性前壁心肌梗死,4周后形成心力衰竭:模型组大鼠尾静脉注射空腺病毒载体,治疗组心力衰竭大鼠尾静脉注射腺病毒介导的β肾上腺素受体激酶抑制剂(Ad-βARKct);另一组为假手术组尾静脉注射空腺病毒载体。4周后测定各组大鼠血儿茶酚胺和NT-ProBNP水平以及左心室非梗死区心肌组织儿茶酚胺水平、β肾上腺素受体激酶(βARK1)以及β肾上腺素受体的表达。结果与模型组比较,治疗组大鼠血NT-ProBNP水平降低(P<0.01),血儿茶酚胺水平降低而心肌组织儿茶酚胺水平升高(均P<0.01),βARK1 mRNA水平降低(P<0.01)且βARK1蛋白表达水平降低(P<0.01),β肾上腺素受体mRNA水平升高(P<0.01)。结论通过尾静脉注射Ad-βARKct可以明显改善心肌梗死后心力衰竭大鼠的神经内分泌激素水平及β肾上腺素受体信号传导。Objective To study the effect of adenovirus-mediated β-adrenergic receptor kinase inhibitor (βARKα) on neuroendocrine hormone and myocardial β-adrenergic receptor signaling system through peripheral vein in a rat model of heart failure after myocardial infarction. Methods The male SD rats weighing about 250 g each were divided into three groups. Two groups underwent ligation of the left anterior descending (LAD) coronary artery to induce acute myocardial infarction, and heart failure occurred 4 weeks later: model group (the heart failure rats were injected empty adenovirus vector though the caudal vein), treatment group (the heart failure rats were injected Ad-βARKα though the caudal vein); and sham-operated group (the rats were injected empty adenovirus vector though the caudal vein). After 4 weeks, Catecholamine and NT-ProBNP of blood plasma, catecholamine and the expression of βARK1 and βAR of non-infarcted left ventricular tissue was measured. Results Compared with the control group, the plasma NT-ProBNP was lower(P〈0.01), the plasma eatecholamine in experimental group was lower, the cateeholamine of myocardial tissue was higher (both P〈0.01), the βARK1 mRN A expression was downregulated (P〈0.01), the βARK1 protein expression Was do wnregulated (P〈0.01), and the β-adrenergic receptor mRNA expression were upregulated (both P〈0.01) in experimental group. Conclusion Ad-βARKct can improve neuroendoerine hormone and β-adrenergic receptor signal transduction in heart failure rats after myocardial infarction.
