摘 要:目的了解EV71病毒传代稳定性,为手足口病的疫苗研究及预防提供参考依据。方法将TZ35号EV71病毒株在Vero细胞连续传20代,对第1代、第20代毒株致细胞病变、感染细胞免疫荧光染色、病毒感染力、VP1基因核苷酸序列及氨基酸序列进行比对分析。结果 TZ35号EV71病毒在Vero细胞中连续传20代,其致细胞病变特点、感染细胞免疫荧光染色特点没有明显改变;VP1基因核苷酸序列没有改变;第1代、第20代CCID50/ml分别为5.5、7.25。结论EV71病毒在Vero细胞中连续传20代,其生物学特性没有明显改变,VP1区基因序列没有改变,提示同一病毒株的遗传是相对稳定的。Objective To understand the genetic stability of EV71 in passage, and provide reference basis for vaccine research and prevention of HFMD. Methods The EV71 TZ35 strain was passaged in Vero cells for 20 generations continuously. The 1 st -generation viral strain and the 20'h -generation strain were compared and analyzed in cell mutagenicity, the immunofluores- cence staining of infected cells, virus infectivity, the nucleotide sequences and amino acid sequence of VP1 gene. Results No significant changes were found in cell mutagenicity and the immunofluorescence staining of infected cells after continuous 20 - generation passage of EV71 TZ35 in Vero cells, and no change was found in the nucleotide sequence of VP1 gene. The CCID50/ml of the 18, generation strain and the 20'h generation strain were 5.5 and 7.25 respectively. Conclusion The biological characteristics and VP1 genetic sequences of EV71 had no significant change after continuous passage in Vero cells for 20 generations, indicating that the inheritance of the same virus was stable.
