摘  要:目的:观察红花黄色素(SY)抑制血小板激活因子(PAF)诱发的内皮细胞炎性因子蛋白表达升高及抑制PAF与其受体特异性结合的作用。方法:用酶联免疫吸附测定法(ELISA)检测SY缓解1μmol/L PAF刺激脐静脉内皮细胞株Eahy926培养液中的炎症因子白细胞介素(IL)-1β、IL-6和肿瘤坏死因子(TNF)-α的蛋白表达水平的变化;以放射配基结合试验观察SY抑制[3H]标记的PAF与家兔血小板膜和膜蛋白PAF受体特异性结合的作用。结果:与正常组比较,PAF损伤组内皮细胞培养上清液中IL-1β、IL-6及TNF-α蛋白表达水平明显升高(P<0.001),SY处理组(浓度为4.5×10-5,4.5×10-4,4.5×10-3g/L)时可明显抑制Eahy926培养液中IL-1β、IL-6及TNF-α蛋白表达水平的升高(P<0.05,0.01,0.001);受体结合实验结果表明浓度为16.49g/L的SY可抑制0.17 nmol/L[3H]PAF与家兔血小板膜的特异性结合,浓度为11.17和22.08g/L的SY分别可抑制0.083及0.25nmol/L[3H]PAF与兔血小板膜蛋白的特异性结合。结论:SY通过抑制PAF与其受体的特异性结合缓解了PAF诱发的血管内皮细胞炎症介质IL-1β、IL-6及TNF-α蛋白的高表达。Objective:To observe the inhibitory effect of samor yellow (SY) on endothelium inflammatory factor proteins expression elevation triggered by platelet activating factor(PAF).Methods:Umbilicus vein endothelium cell line Eahy926 was used.The cell culture supernatants were collected from Eahy926 cells 4 h after SY and PAF addition.The concentrations of IL-1β,IL-6 and TNF-o were measured by ELISA technology.The specific binding of [3H] PAF to rabbit platelet membrane and platelet membrane protein were observed by radio ligand binding assay (RLBA).Results:Endothelium IL-1β,IL-6,TNF-α protein expression levels increased after PAF addition and SY can attenuate these changes.The PAF receptor specific binding inhibition effects of SY were observed.Conclusion:The specific binding of PAF to its receptor can be antagonized by SY and SY is effective to inhibit endothelium IL-1β,IL-6,and TNF-α protein expression elevation induced by PAF.
