摘  要:目的:研究在脂质体Lipofectamine 2000的介导下,293T细胞对介孔二氧化硅纳米材料(mesoporous silica nanomaterials,MSNs)的摄取变化。方法 :制备和表征MSNs;293T细胞分别与100μg/mL MSNs、MSNs+Lipofectamine2000共孵育,通过透射电镜(transmission electron microscope,TEM)观察及定量分析293T细胞对材料的摄取。结果:TEM、X线衍射及氮气吸附-脱附分析表明,制备的MSNs结构规则、分散性好,尺寸在100 nm左右;TEM定量结果发现,Lipofectamine 2000显著提高了293T细胞对MSNs的摄取,P<0.001。结论:Lipofectamine 2000能提高293T细胞对MSNs的摄取,为MSNs在293T细胞内的生物应用创造了条件。MSNs were synthesized and characterized; 293T cells were incubated with MSNs or MSNs and Lipofectamine 2000 re- spectively, which both had a concentration of 100 μ g/mL. Transmission electron microscope (TEM) was used to analyze the up- take of MSNs in 293T cells. Results The analyses by TEM, X-ray diffraction and Nitrogen absorption-desorption showed the prepared MSNs behaved well in structure and dispersion and had a size of 100 nm. TEM quantitative analysis found that Lipo- fectamine 2000 significantly enhanced the uptake of MSNs in 293T cells, with P〈0.01. Conclusion MSNs uptake promoted by Lipofectamine 2000 lays a foundation for its biological application in 293T cells.
