摘 要:目的:研究三叶人字草(Ks)对胶原诱导性关节炎(CIA)大鼠的作用,并初步探讨其抗氧化机制。方法:尾根部皮下注射牛Ⅱ型胶原与不完全弗氏佐剂混合物的方法诱导出CIA大鼠模型,Ks低、中、高3个剂量(4 g、8 g、16 g生药/kg)于第1次免疫后14 d开始灌胃进行干预。在给药后第0、5、10、15、20、25 d对大鼠左右后足关节红肿程度进行关节炎指数评定,28 d后取材,石蜡切片,HE染色观察各组大鼠踝关节组织病理学变化,并检测血清中丙二醛(MDA)含量及总抗氧化能力(T-AOC)和总超氧化物歧化酶(T-SOD)的活性水平。结果:Ks高剂量组能减轻CIA大鼠的关节炎肿胀,降低血清中MDA水平,升高T-AOC、T-SOD活性,具有明显的抗氧化能力。同时病理检查发现,可以改善局部关节炎症状,抑制CIA大鼠滑膜细胞增生,减轻炎性细胞浸润。结论:Ks可以缓解CIA模型的关节肿胀程度,抗氧化作用可能是其作用机制。Objective: To research the impacts of Kummerowia striata (Ks) on collagen-induced arthritis rat and detect its antioxidant mechanism preliminarily. Methods: Collagen- induced arthritis (CIA) rats model were induced by injection with the mixture of bovine Ⅱ collagen and incomplete Freund 's adjuvant on the root of tail. Ks was orally administrated in low, medium or high dosages(4,8,16 g crude drug/kg) beginning 14 days after the first immunization. On days 0, 5, 10, 15, 20, 25 after gavage, the swelling degree of the hind foot was evaluated on arthritis index. On day 28 after gavage, rats were sacrificed for material. Histological analysis of ankle joint was visualized by hematoxylin-eosin (HE) staining, meantime, malondialdehyde (MDA) serum content and total antioxidant capacity (T-AOC)and total superoxide dismutase (T-SOD) serum activities were detected. Results: Ks in the high dosage could relieve the swelling degree of arthritis, and lowered serum MDA contentand elevated T-SOD and T-AOC serum activities, which showed the obvious antioxidant capacities. Meanwhile, local symptoms of arthritis were alleviated, such as inhibiting synovial cell proliferation and inflammatory cell infiltration. Conclusion: Ks could relieve the joint swelling of CIA rat, which might associated with enhancing antioxidant capacities.
