益母草注射液有效部位提取及其对凝血系统的影响研究

摘　　要：目的:探索益母草注射液对凝血功能的影响及其有效物质基础。方法:首先对益母草注射液进行提取分离,然后通过体外实验测定凝血4项值、血小板聚集率和纤溶活性,探索益母草注射液及其提取物对凝血系统的影响。结果:凝血4项值测定结果表明,益母草注射液水溶性非生物碱部分体外给药能明显延长PT、APTT,益母草注射液水溶性生物碱部分体外给药能明显缩短APTT,两者均明显降低血纤维蛋白原含量和明显延长TT。血小板聚集实验表明,益母草注射液两大部分均具有明显的抗血小板聚集活性,尤以水溶性非生物碱部分的抗血小板聚集活性更强,且呈明显的量效关系。纤溶实验表明,益母草注射液水溶性非生物碱部分具有明显的纤溶活性且呈明显的量效关系。益母草注射液水溶性生物碱部分未见明显的纤溶活性。结论:益母草注射液同时具有活血与止血的作用,止血作用机制与缩短内源性凝血时间有关,活血作用机制与延长内外源性凝血时间、抗血小板聚集和纤维蛋白溶解有关。益母草注射液水溶性非生物碱部分是益母草注射液活血化瘀的主要部分,该提取分离方法对益母草注射液质量控制研究具有重要意义。Objective： To explore the effects of Motherwort injection and its effective parts on coagulation system. Methods： At first, extract and separate motherwort injection, then test the effects of effective parts on four items of blood coagulation values, platelet aggregation and fibrinolysis in vitro. At last, we can find out the effects of motherwort injection and its extract on blood coagulation system. Results： Coagulation values measured results show that Motherwort injection water-soluble non-alkaloids administrated in vitro could significantly prolong the PT, APTT, motherwort injection soluble alkaloids administrated in vitro can significantly shorten the APTT, both significantly reduce blood fibrinogen levels and prolonged TT. Platelet aggregation experiments showed that the two parts from motherwort injection obviously have antiplatelet aggregation activities, especially the water-soluble non-alkaloid. It also showed a clear dose-effect relationship. Fibrinolysis experiments show that motherwort injection water soluble non-alkaloids has significant fibrinolytie activity, and showed a clear dose-response relationship. Motherwort injection soluble alkaloids showed no fibrinolytic activity. Conclusion： Motherwort injection contains both anticoagulant and procoagulant ingredients. Motherwort injection watersoluble non-alkaloid part is the main part with anticoagulant. The methods of extraction and separation are great significant for the quality control of motherwort injection.
