藤龙补中汤对晚期大肠癌患者调节性T细胞作用临床研究

摘　　要：目的大肠癌是全球常见恶性肿瘤;调节性T细胞(Regulatory T cells,Tregs)是一群免疫负调控细胞,参与大肠癌的发病,与大肠癌预后直接相关。藤龙补中汤是以解毒健脾立法的中药复方,细胞与动物模型研究显示藤龙补中汤可以促大肠癌细胞凋亡、细胞衰老,增强化疗对大肠癌的疗效。本研究观察了藤龙补中汤对晚期大肠癌患者Tregs的作用。方法晚期大肠癌患者随机分为对照组及治疗组,分别给化疗或藤龙补中汤联合化疗治疗;观察患者治疗前后Tregs细胞及相关细胞因子变化。流式细胞仪检测Tregs细胞,酶联免疫吸附实验(Enzyme-Linked Immunosorbent Assay,ELISA)检测细胞因子。结果治疗组治疗后调节性T细胞及白介素-10(Interleukin-10,IL-10)、转化生长因子-β(Transforming growth factor-β,TGF-β)水平显著降低(P<0.01),与对照组比较差异显著(P<0.01)。结论藤龙补中汤可以降低晚期大肠癌患者Tregs细胞数量,以相关细胞因子IL-10、TGF-β水平。Objective Colorectal cancer is the common malignant tumor in the world.The regulatory T cells（Tregs）are the group of immune negative regulatory cells,involved in the occurrence of colorectal cancer and closely related to the prognosis of colorectal cancer.Tenglong Buzhong Decoction is the compound Chinese medicine for removing toxin and strengthening the spleen.The cell and animal model study showed that Tenglong Buzhong Decoction promoted colorectal cancer cell apoptosis and call aging and enhanced the effect of chemotherapy for colorectal cancer.The study observed the effect of Tenglong Buzhong Decoction on Tregs in the patients of late colorectal cancer.Methods The patients of late colorectal cancer were random-ized into a control group and a treatment group,treated with chemotherapy or the combined therapy of Ten-glong Buzhong Decoction and chemotherapy（combined therapy）separately.Tregs and the relevant cytokines were observed before and after treatment in the treatment.Flow cytometry（FCM）was used to detect Tregs. Enzyme-linked immunosorbent assay（ELISA）was to detect cytokines.Results In the treatment group,the levels of Tregs,interleukin-1 0（IL-1 0）and transforming growth factor-β（TGF-β）were reduced signifi-cantly after treatment（P〈0.01 ）and were different significantly as compared with those in the control group （P〈0.01 ）.Conclusion Tenglong Buzhong Decoction reduces the count of Tregs and the levels of IL-1 0 and TGF-βin the patients of late colorectal cancer.
