摘 要:为寻找对豁眼鹅部分生产性状进行选育的有效分子标记,根据鹅GH基因的cDNA序列设计引物,利用PCR-SSCP技术对饲养于辽宁省和江苏省的豁眼鹅(共计164只,同批出雏)GH基因外显子2进行SNPs检测,并克隆测序其纯合基因型,与10周龄的屠体和肉品质性状进行关联分析。结果表明:GH基因外显子2共有2个SNPs突变位点,得到AA、BB、CC、DD、AB、AC、AD、BC、BD和CD 10种基因型,将氨基酸序列突变和未突变的基因型各自合并后得到的3种基因型;外显子2的部分基因型对部分屠体性状均有极显著(P<0.01)或显著(P<0.05)影响,对部分肉品质性状有显著影响(P<0.05);在公鹅和母鹅群体中,AB、BB基因型分别为部分屠体性状和肉品质性状的优势基因型,可以用于对豁眼鹅生产性能选育的进程中。In this study one primer for exon 2 in growth hormone (GH) gene was designed according to the cDNA sequence of GH gene of goose, and single nucleotide polymorphism (SNP) of exon 2 in GH gene in Huoyan goose (atotal of 164 individuals) were detected by PCR-SSCP method. And homozygotes were subsequently cloned, sequenced and analyzed. 2 SNPs mutations were detected and 10 genotypes including AA, BB, CC, DD, AB, AC, AD, BC, BD and CD were obtained. Correlation analysis of the 3 genotypes which were got after the combination of the 10 genotypes based on whether the sequence of amino acid residues changed or not and carcass and meat quality traits were processed by SPSS13.0 at 10 weeks. The results showed that partial carcass and meat quality traits were extremely different (P〈0.01) or different (P〈0.05) among different genetypes. In male and female populations, AB or BB were high productive genotypes which could serve as molecular markers for the breeding and selection of carcass and meat quality traits in Huoyan goose.
