摘  要:本研究为提高黄霉素的发酵效价及发酵稳定性,对加纳链霉菌种子培养工艺、生长特性及移种标准进行研究。结果表明:斜面移种标准为平板单菌落移种第一代斜面,菌龄8d;种子摇瓶培养的最佳培养条件为采用具挡板摇瓶、温度34℃、pH7.0;液体种子移种标准为菌浓35%-41%,pH值二次回落6.9-7.0,还原糖28.8--30g/L,氨态氮二次回落1.05~2.57mg/L,种龄44~48h。摇瓶实验结果显示,使用最佳培养工艺及移种标准,黄霉素的最终效价提高了32.5%.不N批次间的差异低于7%。该实验为提高黄霉素发酵效价和发酵稳定性提供参考。The study was aimed to improve the fermentation pot ency and fermentation stability. The seed culture conditions, growth characteristics and inoculation standards of Streptomyces ghanaensis were studied in shake flask. The results showed that the cultivation of single colony on the plate for 8 days were the best inoculation standard of the slant strains, the optimized culture conditions of liquid seed were established in shake flask with baffles, culture temperature at 34~C, initial pH of 7.0. The inoculation standards of liquid seed consisted of cell eoneetration(35%~41%), pH(6.9- 7.0 after the second drop), reduced sugar (28.8-30 g/L), amino nitrogen (1.05-2.57 mg/L after the second drop) and inoculum age (44-48 h). Using optimal seed culture technology and inoculation standard, the final titer of flavomycin was improved by 32.5%, and the divergence between different batches was less than 7%. This experiment would be helpful in improving flavomycin production and fermentation stability.
