摘 要:目的:建立测定复方降脂颗粒中丹酚酸B和大黄酚含量的方法。方法:采用高效液相色谱法。色谱柱为Agilent Eclipse XDB C18(250mm×4.6mm,5μm),流动相分别为甲醇一乙腈-0.1%甲酸溶液(25:10:65,纠阿y)、乙腈一0.1%磷酸溶液(梯度洗脱),流速为1.0ml/min,检测波长分别为286、284nm。结果:丹酚酸B和大黄酚的进样量分别在1.91~19.12、0.0144~0.1440gg范围内与各自峰面积积分值呈良好的线性关系(r分别为0.9998、0.9999);二者精密度、稳定性、重复性试验的RSD〈2%;平均加样回收率分别为98.58%(RSD=0.97%,n=9)、97.71%(RSD=0.59%,n=9)。结论:该方法简单、准确、重复性好,可用于复方降脂颗粒的质量控制。OBJECTIVE: To establish a method for the content determination of salviamolic acid B and chrysophanol in Compound jiangzhi granules. METHODS: HPLC method was adopted. The determination was performed on Agilent Eclipse XDB C18 (250 mm×4.6 mm, 5 μm) column with mobile phase consisted of methanol-acetonitrile-0.1% formic acid (25: 10:65, V/V/V) for salviamolic acid B and acetonitrile-0.1% phosphoric acid for chrysophanol (gradient elution) at the flow rate of 1.0 ml/min. The detection wavelength were 286 mn for salviamolic acid B and 284 nm for chrysophanol. RESULTS: The linear ranges were 1.91-19.12 μg for salviamolic acid B (r=0.999 8) and 0.014 4-0.144 0 μg for chrysophanol (r=0.999 9); RSDs of precision, sta- bility and reproducibility tests were all lower than 2% with an average recovery of 98.58% (RSD=0.97% , n=9) and 97.71% (RSD=0.59%, n=9). CONCLUSIONS: The method is simple, accurate and reproducible, and can be used for quality control of Compound jiangzhi granules.
