摘  要:目的:研究复方丹参注射液对麻醉犬急性心肌缺血的保护作用。方法:健康杂种犬30只随机分为模型对照组、复方丹参注射液低、中、高剂量组和阳性药物香丹注射液对照组,每组6只。采用结扎冠状动脉前降支的方法制备犬急性实验性心肌缺血模型。检测犬心肌氧摄取率(VO2)、心肌耗氧量(MVO2)、氧分压(PO2)、血氧含量与动、静脉血氧含量差;心外膜电图描记和硝基蓝四唑(N-BT)组织化学染色方法计算心肌缺血程度及梗死面积;应用生化自动分析仪测定血清乳酸脱氢酶(LDH)和肌酸磷酸激酶(CPK)。结果:复方丹参注射液能降低犬心肌耗氧量和氧摄取量,增加动静脉氧差,明显减轻由心外膜电图标测的心肌缺血程度,缩小经N-BT染色所显示的梗死区,显著增加缺血心脏的心肌血流量,对心肌缺血及心肌梗死引起的血浆LDH和CPK的含量升高有明显抑制作用。结论:复方丹参注射液对犬急性心肌缺血和心肌梗死有明显的保护作用,可能通过抑制LDH和CPK的释放,增加心肌血流量等机制完成。Objective:To study the protective effects of compound Danshen injection on acute myocardial ischemia in anaesthetic dogs.Methods:The acute ischemia models were made by ligation of left anterior descending (LAD) artery.VO2,MVO2,oxygen contents and oxygen content difference were measured,epicardiogrammapping was adopted to measure the scope and degree of myocardial ischemia,quantitative histologic assay (nitroblue tetrazolium,N-BT stain) to determine the size of myocardial infarction,and the lactate dehydrogenase (LDH) and creatine phosphokinase (CPK) contents were measured by automatic biochemistry analyzer.Results:Compound Danshen injection obviously alleviate the degree of myocardial ischemia (Σ-ST) and reduce the ischemic area indicated by N-BT staining; in addition,compound Danshen injection could increase the MBF of ischemia section and inhibit the LDH and CPK release induced by ischemia and infarction.Conclusions:Compound Danshen injection demonstrated to attenuate the damage subjected to myocardial ischemia and infarction,which may be due to its function of inhibiting the LDH and CPK release,increasing the MBF,and then improving the damaged cardiac function.
