摘  要:目的:建立一种简便且准确测定槟榔中总生物碱含量的方法。方法:以氢溴酸槟榔碱为对照品,采用酸性染料比色法测定总生物碱的含量,测定波长为618 nm。结果:槟榔总生物碱在20~120μg呈线性关系,回归方程为Y=4.226 7 X-0.005 2(r=0.999 6),平均回收率为99.6%,RSD=1.54%。稳定性、精密度和重复性都比较高,RSD分别为1.32%,0.109%,2.18%。样品分析结果为红心槟榔含量最高,常见槟榔含量次之,西瓜槟榔较低。结论:该方法操作简便、灵敏、准确,可用于槟榔总生物碱的质量控制。Objective: To establish a simple and accurate method for the determination of total alkaloids in arecanut. Methods: The determination was carried out by acid dye colorimetry with arecoline hydrobromide as reference, at wavelength of 618 nm on UV-2450 spectrophotometer. Results: The total alkaloid contents of arecanut were linear in the range of 20 to 120 p^g, the regression equation was Y = 4. 226 7X -0. 005 2 (r =0. 999 6) , the average recovery was 99.6% with a RSD of 1.54%. There was a relatively high stability, precision and repeatability, RSD were 1.32%, 0.109% and 2.18% respectively. The results of sample analysis showed the highest total alkaloid content was in red arecanut germplasm, followed by common arecanut germplasm, the lowest in watermelon arecanut germplasm. Conclusion: The method is simple, sensitive and accurate.
