摘 要:目的:建立测定异黄酮类化合物BI的含量及有关物质的高效液相色谱方法。方法:采用AgilentZorbaxescilpseXDBC18(250mm×4.6mm,5.0μm)色谱柱;以0.15%醋酸-甲醇(45:55)为流动相;流速:1.OmL/min;检测波长为260nm;柱温40℃;进样体积:10μL。结果:在上述色谱条件下,各杂质及各降解产物均可与BI主峰良好分离,HPLC测定的线性范围为0.08μg-0.80μg(r=0.9999),方法的仪器精密度、日内精密度、日间精密度及重复性的RSD均小于2%。结论:采用HPLC测定中药BI的含量及有关物质,方法简便,结果准确可靠。Objective:To establish an RP-HPLC method for determination of the content and the related substances of Isoflavone compounds,BI.Methods:The separation was performed on an Agilent Zorbax escilpse XDB C18 column (250mm ×4.6mm,5.0μm);the mobile phase consisted of methanol-0.15%acetic acid (55:45) at the flow rate of 1.0 mL/ min;the detective wavelength was set at 260nm;the column temperature was 40 ℃;and the injection volume was set at 10μL.Results:An excellent separation was achieved for BI and its related substances.The assay was validated in the concentration range of 0.08μg-0.80μg (r=0.9999).All of the instrument precision,precision in a day, precision during the days and the repeatability were less than 2%.Conclusion:The developed method can give more accurate and sensitive results for the determination of the content and the related substances of BI.
