摘  要:目的探讨阳离子脂质体介导的A20基因转染人外周血单核细胞的可行性及最佳转染条件。方法采用脂质体Lipofectamine TM 2000包裹pCAGGS-GFP/A20质粒转染人外周血单核细胞,通过荧光显微镜检测GFP报告基因,RT-PCR检测A20基因表达。结果应用Lipofectamine TM 2000介导的A20基因转染人外周血单核细胞,在2×106/L细胞接种浓度,脂质体与质粒的量之比为4μl∶3μg时,转染效果最佳,RT-PCR证实成功获得A20目的基因的表达。结论脂质体转染法可介导A20基因在人外周血单核细胞获得表达,该研究为A20基因转染的相关实验研究提供参考依据。Objective The feasibility and the optimal transfection conditions of human peripheral blood mononuclear of cationic lipo- some mediated A20 gene transfeetion were investigated in this paper. Methods Human peripheral blood mononuclear cells were trans- fected by pCAGGS - GFP/A20 plasmid encapsulated by liposome Lipofectamine TM 2000, GFP reporter gene was detected by fluorescence microscopy expression of A20 gene was detected by RT - PCR. Results Transfectiun effeet was hest when cells inoculation concentration was 2 x 106/1, and liposome and plasmid ratio was 4μl: 3μg by applying Lipofectamine TM 2000 mediated A20 gene to transfeet blood mononuelear cells of human peripheral. RT - PCR confirmation successfully achieved the expression of A20 objective gene. Conclusion The method of liposome transfection mediated A20 gene in human peripheral blood mononuciear cells was expressed. Such study provides a reference for related experimental study of A20 gene transfection.
