摘 要:目的观察模拟海拔5000米高原急性缺氧条件下大鼠肾脏氧化应激及NADPH氧化酶亚基表达的时程变化,探讨急性缺氧对肾损害的分子机制。方法采用低压氧舱复制大鼠急性缺氧模型,设置0.5、1、3、5、7天,5个不同缺氧时间点,同时设置对照组(西宁地区),于不同时间取大鼠各10只取材,采用生化分析仪测定大鼠血液中肌酐及尿素氮水平,采用比色法测定大鼠肾脏中超氧化物歧化酶(SOD)、丙二醛(MDA)含量,采用Western blot方法,测定大鼠不同缺氧时间组肾脏NOX4及P47phox蛋白表达变化。结果随着缺氧时间的延长,大鼠肌酐及尿素氮水平呈先升高后下降的趋势;肾脏SOD水平则一直下降,差异具有统计学意义(P<0.05),MDA表达水平总体呈升高趋势,但差异无统计学意义(P>0.05);大鼠肾脏NOX4及P47phox蛋白表达量随着缺氧时间延长而开始升高,于5天后开始降低,7天恢复大致正常水平。结论急性缺氧早期肾脏NOX4及P47phox蛋白表达水平明显增多,在缺氧所致的肾脏氧化应激对早期肾脏损伤起重要作用。Objective To observe the expression of the rat renal oxidative stress and NADPH oxidase suhunit along with the sched- ule changes using simulating the plateau low oxygen environment at an altitude of 5000 meters. Methods Using low pressure oxygen tank to duplicate the model of the rats with acute hypoxia, set up 0.5 day, 1 day, 3 day, 5 day, 7 day, five different oxygen point in time, at the same time set the control group (Xining area). Samples were taken at different time using each 10 rats using biochemical analyzer to determine rats creatinine and urea nitrogen levels in the blood. The kidney of rats was evaluated by colorimetric method to determine the super oxide dismutase (SOD) , malondialdehyde (MDA) content. Using Western blot method to determine the rats kidney NOX4 and P47phox protein expression changes at different oxygen time. Results Along with the duration of hypoxia, rats creatinine and urea nitro- gen levels showed a trend of rise after the fall at first, renal SOD level has been falling all the time. there were obvious differences in both groups (P 〈 0.05). MDA expression level rising all through, but there was no significant difference (P 〉 0.05 ). Rat kidney NOX4 and P47phox protein expression began to rise with prolonged hypoxia time, after 5 days it began to reduce, after 7 days it renewed at the rough- ly normal levels. Conclusion Kidney NOX4 and P47phox protein expression level were increased significantly at the acute hypoxia early phase. Hypoxia causing the renal oxidative stress plays an important role in early renal damage.
