摘 要:目的探讨和分析加巴喷丁(GBP)对长春新碱(VCR)致神经病理性疼痛大鼠大脑皮层蛋白表达的影响和可能的中枢机制。方法 36只成年SD大鼠随机分为3组:空白对照组(Ns组)给予生理盐水,疼痛模型对照组(Pc组)给予VCR,GBP治疗组(Gt组)给予VCR、GBP。给药结束后,光镜和电镜观察各组大鼠坐骨神经超微结构;双向凝胶电泳(2-DE)各组脑皮层组织蛋白质,质谱鉴定并分析差异表达的蛋白质。结果 (1)光镜和电镜下坐骨神经超微结构:Ns组为正常大鼠坐骨神经纤维结构,Pc组明显退行性变,Gt组变性较Pc组轻微。(2)双向凝胶电泳串联质谱结果:Gt组与Pc组比较,差异表达的蛋白点15个,肽质量指纹图谱鉴定出9种蛋白质,其中5种DJ-1、ATP合成酶亚基D、血红蛋白A链、血红蛋白B链、Cu-Zn超氧化物歧化酶表达上调;4种磷脂酰乙醇胺结合蛋白-1、过氧化物酶-2、核苷二磷酸激酶A、热休克蛋白10表达下调。结论 GBP对VCR致神经病理性疼痛大鼠大脑皮层蛋白质表达存在影响,可能存在中枢层面蛋白水平机制。Objective By detecting differentially expressed proteins in rat cerebral cortex with two - dimensional gel electrophoresis (2 - DE) and tandem mass spectrometry (MALDI -TOF/TOF - MS) techniques, to explore and ana-lyze the impact of gabapentin (GBP) on vincristine (VCR) - induced neuropathic pain as well as the possible mecha- nism. Methods Thirty - six SD rats were randomly divided into 3 groups : blank control group ( Group Ns, saline), pain model control group (Group Pc, VCR) and gabapentin treatment group (Group Gt, VCR and GBP). The ultrastruc-ture of sciatic nerve was observed with light microscope and electron microscope. The differentially expressed proteins in cortex tissue of rat were separated by 2 - DE, and subsequently identified by MALDI - TOF/TOF - MS. Results Normal sciatic nerve fiber structure was observed in Group Ns, while it was markedly degenerated in Group Pc, which was partly reversed in Group Gt. Compared with Group Pc, 15 differentially expressed proteins were spotted in Group Gt, among which 9 proteins were identified. Among them, up - regulation was observed in DJ - 1, ATP synthase subunit D, A chain of hemoglobin, B chain of hemoglobin and Cu - Zn superoxide dismutase ; while down - regulation was observed in phos- phatidylethanolamine binding protein - 1, peroxide enzyme - 2, nucleoside diphosphate kinase A and heat shock protein - 10. Conclusion Gabapentin affects the expression of cerebral cortex proteins in vincristine - induced neuropathic pain model.
