摘  要:观察黄芪注射液(AI)对地塞米松(Dex)所致免疫功能低下小鼠脾淋巴细胞凋亡与增殖的影响,并探讨其作用机制。将72只昆明小鼠随机均分为对照组,模型组,黄芪注射液高、中、低剂量组(2.0、1.0、0.5g/kg),白细胞介素-2(IL-2)组(5IU/kg),除对照组外,其余小鼠均采用Dex(25mg/kg)腹腔注射复制小鼠脾脏淋巴细胞凋亡模型。观察黄芪注射液对小鼠脾脏指数的影响及脾脏的形态学改变,采用免疫组织化学法检测小鼠脾脏淋巴细胞天冬氨酸特异性的半胱氨酸蛋白酶-3(Caspase-3)与增殖细胞核抗原(PCNA)的表达。结果表明,黄芪注射液能明显改善萎缩的脾脏,增加脾脏指数(P<0.05);黄芪各剂量组小鼠脾脏皮质细胞明显增多;同时发现黄芪注射液能降低Caspase-3的表达,并能提高PCNA的表达(P<0.05,P<0.01)。说明黄芪注射液有明显的抗脾脏萎缩及抑制脾脏淋巴细胞凋亡的作用,其作用机制可能与下调Caspase-3及上调PCNA的基因表达相关。To study the effects of astragalus injection(AI) on promoting spleen lymphocyte apoptosis and proliferation in immunodeficient mice by dexamethasone(Dex), 72 KM mice were randomly divided into six groups: control group, model group, interleukin-2 group(5 IU/kg), high AI group(2 g/kg), middle AI group(1 g/kg) and low AI group(0.5 g/kg). Besides control group,the mice in other five groups were injected Dex 5 days for immunodeficient mouse model. The effects of AI on the spleen index and morphological changes in the spleen were observed,Casepase-3 and PCNA were detectedby immunohistochemistry. AI can significantly improve the atrophy of the spleen, and increase spleen index (P〈0.05);AI can significantly increase cortical cells of the spleen and splenic lymphocytes;Also AI can reduce the expression of Caspase-3, while improve the expression of PCNA (P〈0.05, P〈0.01). AI has significant anti-spleen atrophy and inhibition of apoptosis of spleen lymphocytes, and its mechanism may be by down Caspase-3 and up PCNA related-gene expression.
