摘  要:细胞融合技术是细胞工程中的一项核心基础技术,聚乙二醇(PEG)是最常用的促融剂,广泛地被应用于农业、医药、环保等多个领域。探讨更符合细胞工程要求的、融合后的细胞更大限度地保持生理活性的、快速、有效的细胞融合条件。以牛红细胞为实验材料,PEG作促融剂介导牛红细胞融合,研究不同的融合温度、PEG作用时间、PEG的相对分子质量和PEG的质量分数等因素对细胞融合率的影响。实验结果表明,选择融合温度为39℃,时间为15min,选用相对分子质量为2000、质量分数为45%的PEG,可以在细胞受毒害较小的同时得到最佳的融合效果,融合率能达到21.26%.The cell-fusion technique is a core basic technology ,as the most frequently revulsant ,PEG is widely used in many fields such as agriculture、medicine、environmental protection,and so on.To find a more effective and rapid cell -fusion meth-od that can answer for the cell -engineering needs which need the fused cells to keep more physiological activities better , here, ox red cells were taken as the material , which were made to fuse induced by PEG, the author studied the effects of fu-sion temperature、PEG role time、PEG molecular weight and mass fraction on cell fusion .The results showed that when the fu-sion temperature、PEG role time、PEG molecular weight and mass fraction was 39℃、15min、2000、45%,respectively, the fu-sion frequency reached to the peak value at the same time that the poison to cells was much less ,and the cell fusion rate reached to 21.26%.
