摘  要:为了研究动物杂种优势的分子遗传机制,研究以北京鸭、法国番鸭及其正反交F1代杂种为材料,采用mRNA差异显示(DDRT-PCR)方法分析不同试验组鸭肌肉组织蛋白L异天冬氨酸(D-天冬氨酸)O-甲基转移酶1(PCMTD1)基因的表达差异,结合抑制性消减杂交(SSH)方法加以验证。结果表明:PCMTD1基因在杂种中表达上调;与鸭PCMTD1基因3'-UTR 795 bp片段进行比对,与鸡(Gallus gallus)PCMTD1同源性为85%,期望值(E值)为0。说明杂种优势相关基因克隆成功。To investigate the molecular genetic mechanism of animal heterosis, Beijing duck, France muscovy duck and the their mutual hybrids were used as materials to analyze the differential expressions of protein -L - isoaspartate ( D - aspartate) D - methyhransferase domain containing (PCMTD1) genes in duck muscle tissues of different test groups using mRNA differential display (DDRT- PCR). And the display pattern was verified by the method of suppression subtractive hybridization (SSH). The results showed that the expression of PCMTD1 gene was up - regu- lated in the hybrids. The comparison results of the 795 bp fragment of duck PCMTD1 gene 3' - UTR showed that the duck PCMTD1 gene shared 85 percent homology with that of chicken, and the E - value was 0. The results indicate that the gene associated with heterosis is coloned successfully.
