摘  要:目的:观察羌活地黄汤含药血清对胶原性关节炎(CIA)大鼠成纤维样滑膜细胞凋亡的影响,探讨羌活地黄汤抑制滑膜增生的机制。方法:分别予新西兰大白兔灌胃羌活地黄汤、独活寄生汤,制备药物血清。选择10只雌性SD大鼠,6只建立牛Ⅱ型胶原诱导的大鼠关节炎模型,分离培养成纤维样滑膜细胞,分为模型组、羌活地黄汤组和独活寄生汤组;另4只作为正常组(用等量生理盐水)。正常组、模型组细胞均加入5%DMEM血清的培养液;羌活地黄汤组和独活寄生汤组,分别加入含10%相应含药血清的DMEM培养液。采用MTT法检测滑膜细胞增殖,ELISA法检测IL-17、核因子κB受体活化因子配体(RANKL)表达浓度。结果:羌活地黄汤含药血清可抑制滑膜细胞的增殖,与模型组相比差异有统计学意义(P<0.05);同时羌活地黄汤还可抑制IL-17、RANKL的表达(P<0.05)。结论:羌活地黄汤治疗类风湿关节炎的作用机制可能与抑制成纤维样滑膜细胞的增殖及降低IL-17、RANKL的表达有关。Objective : To observe the effect of "Qianghuo Dihuang Decoction" on fibroblast-like synovioeytes of rats with collagen-induced arthritis (CIA) and to explore its mechanism. Methods: Six white rabbits were intragastrically administered with "Qianghuo Dihuang Decoction" and "Dnhno Jisheng Decoction" for 15 days. Six male SD in 10 rats were induced by Type II collagen to establish the models and divided into 3 groups: model group, "Qianghuo Dihuang Decoction" and "Duhuo Jisheng Decoction", and the other 4 rats in normal group were treated with the same volume of normal saline. We isolated and cultured fibroblast-like synoviocytes. Added nutrient solution of 5% DMEM in normal group and model group; the 10% drug serum were added in "Qianghuo Dihuang Decoction" group and "Duhuo Jisheng Decoction" group respectively. Synovioeyte proliferation was determined by MTT assay, and expressions of interleukin- 17 ( IL- 17 ) and receptor activator of NF-κB ligand (RANKL) were measured by ELISA. Results: "Qianghuo Dihuang Decoction" contained serum can restrain the proliferation of synoviocytes, with significant difference between the model group ( P 〈 0.05 ). "Qianghuo Dihuang Decoction" can restrain the expressions of IL- 17 and RANKL ( P 〈 0.05 ). Conclusion: The mechanism of "Qianghuo Dihuang Decoction" may be association with restraining the proliferation of synoviocytes and inhibiting the expression of IL-17 and RANKL.
