摘  要:[目的]使用低温定向冷冻方法建立有效的精子冻存条件。[方法]采用低温定向冷冻方法冻存食蟹猴精液,并与常规方法进行对比,探讨冷冻方法和甘油浓度对精子冷冻存活率的影响。[结果]在相同甘油浓度下,低温定向冷冻的食蟹猴精子的活力与顶体完整性显著高于常规冷冻法(P<0.05),定向冷冻方法在保存食蟹猴精液效果明显优于常规冷冻方法,且甘油浓度5%时能够达到更好的冷冻效果。[结论]与常规冷冻相比,低温定向冷冻能显著提高精子存活率。在食蟹猴的精液冻存中,采用定向冷冻仪冻存精液能达到较好的冻存效果。[Objective] The research aimed to establish the effective preservation method of frozen sperm by directional freezing technique.[Method] The sperm of M.fascicularis was frozen by directional freezing technique and it was compared with conventional freezing technique.The effects of freezing method and glycerol concentration on the freezing survival rate of sperm were discussed.[Result] At the same concentration of glycerol,The sperm' s motility and acrosome integrity of M.fascicularis by directional freezing technique were higher than that by conventional freezing technique.The preservation effects of sperm of M.fascicularis by directional freezing technique were obviously better than that by conventional freezing technique.And the best freezing effects were obtained with 5% glycerol.[Conclusion] Compared with conventional freezing technique,directional freezing technique could enhance the survival rate of sperm.Better freezing effects were obtained when the sperm of M.fascicularis was frozen by using directional freezing instrument.
